GFR-Vivo 680 Fluorescent Imaging Agent for Toxicology - Application note

چکیده

Glomerular filtration rate (GFR) is the gold standard in kidney function assessment and is used to determine progression of kidney disease and drug-induced kidney toxicity. One of the most accepted ways to assess GFR is by measuring the rate of disappearance of labeled inulin from the blood; as inulin is completely filtered at the kidneys’ glomeruli (but neither secreted nor reabsorbed by the tubules), this rate of disappearance is directly proportional to GFR. We have developed a near infrared (NIR) fluorescent-labeled form of inulin (GFR-VivoTM 680) in a spectral region providing low background and high tissue penetration (ex/em = 670/685 nm) for in vivo application. Fluorescence molecular tomographic (FMT) imaging of the heart was used to detect and quantify blood levels of GFR-Vivo 680 at multiple time points, providing the necessary data to calculate the clearance rates in individual animals. Following an intravenous bolus of NIR-Inulin in SKH-1E mice, FMT® images were acquired at 1, 5, 15, 30, and 45 minutes post-injection. Clearance rates were calculated using a two-compartment curve fitting, yielding average rates of 270 + 6 mL/min in normal mice. GFR-Vivo 680, in combination with FMT heart imaging, provides a non-invasive fluorescent imaging approach to generate consistent GFR measurements in models of kidney disease, dysfunction, and drug toxicity. Evaluation of a 99mTc-tricine Vascular Disrupting Agent as an In-vivo Imaging in 4T1 Mouse Breast Tumor Model

Colchicine as a vascular disrupting agent creates microtubule destabilization whichinduces vessel blockage and consequently cell death. Accordingly, colchicines and itsanalogues radiolabeled with 99mTc may have potential for visualization of tumor. In this work, deacetylcolchicine a colchicine analogue was labeled with 99mTc via tricine as a coligandand characterized for its tumor targeting prop...

Synthesis, Radiolabeling, and Biological Evaluation of Peptide LIKKPF Functionalized with HYNIC as Apoptosis Imaging Agent

Introduction: A noninvasive method of detecting exposure of phosphatidylserine (PS) on the external surface of the plasma membrane such as nuclear imaging could assist the diagnosis and therapy of apoptosis related pathologies. The most studied imaging agent for apoptosis is Annexin V so far. Because of limitations of Annexin V other agents have been introduced such as small peptides and molecu...

Pre-clinical toxicity evaluation of MB-102, a novel fluorescent tracer agent for real-time measurement of glomerular filtration rate.

The fluorescent tracer agent 3,6-diamino-2,5-bis{N-[(1R)-1-carboxy-2-hydroxyethyl]carbamoyl}pyrazine, designated MB-102, has been developed with properties and attributes for use as a direct measure of glomerular filtration rate (GFR). In comparison to known standard exogenous GFR agents in animal models, MB-102 has demonstrated an excellent correlation. A battery of toxicity tests has been com...

Ex Vivo and In Vivo Noninvasive Imaging of Epidermal Growth Factor Receptor Inhibition on Colon Tumorigenesis Using Activatable Near-Infrared Fluorescent Probes

BACKGROUND Near-infrared fluorescence (NIRF) imaging combined with enzyme-activatable NIRF probes has yielded promising results in cancer detection. OBJECTIVE To test whether 3-dimensional (3-D) noninvasive in vivo NIRF imaging can detect effects of epidermal growth factor receptor (EGFR) inhibitor on both polypoid and flat tumor load in azoxymethane (AOM)-induced colon tumors or tumors in Ap...
