چکیده

Specific Aims SA1. Determine which of the seven identified Drosophila caspase genes are expressed in Drosophila melanogaster S2 cells. SA2. Identify which of the expressed caspases are downstream of DIAP1 in the apoptosis pathway of S2 cells. SA3. Explore the order in which expressed initiator and effector caspases are activated and study the function of proteasomal degradation in regulating the initiator caspase DRONC.

DrICE resurrects Grim to antagonize DIAP1

Caspases (cysteine proteases) have been studied extensively over the past 2 decades for their roles in mediating cell death in multicellular organisms. They are typically activated as part of a proteolytic cascade, in which an initiator caspase is recruited, through its association with an adaptor protein, to a large multimeric caspaseactivating complex. Human caspase-9 and its Drosophila homol...

DIAP1Mediated Apoptosis in Drosophila S2 Cells Relies on the Caspases DRONC, DRICE, and DCP1

The caspase cascade in Drosophila is controlled primarily by DIAP1 (Drosophila inhibitor of apoptosis), which exerts its regulatory effect by targeting the initiator caspase DRONC for proteasomal degradation. Proapoptotic signaling stimulates autoubiquitination by DIAP1 and frees DRONC to activate the caspase cascade. We used RTPCR for expression profiling of Drosophila S2 cells and determin...

Drosophila IAP1-Mediated Ubiquitylation Controls Activation of the Initiator Caspase DRONC Independent of Protein Degradation

Ubiquitylation targets proteins for proteasome-mediated degradation and plays important roles in many biological processes including apoptosis. However, non-proteolytic functions of ubiquitylation are also known. In Drosophila, the inhibitor of apoptosis protein 1 (DIAP1) is known to ubiquitylate the initiator caspase DRONC in vitro. Because DRONC protein accumulates in diap1 mutant cells that ...

Drosophila Apoptosis and the Regulation of the Caspase Cascade

The caspase cascade in Drosophila is controlled primarily by DIAP1 (Drosophila inhibitor of apoptosis), which exerts its regulatory effect by ubiquitinating the caspase DRONC, thus targeting it for proteasomal degradation. Proapoptotic signaling stimulates auto ubiquitination by DIAP1 and frees DRONC to activate downstream caspases.

A Protein Complex Restrains a Homicidal Enzyme during Sperm Differentiation

If one were to take a moral view of cell activities, the caspases would be condemned on two counts. They not only give the order to kill cells, they also carry it out. But in the real-life utilitarian world of the cell, all this killing is simply a means to an end. Programmed cell death, or apoptosis, plays an important role in embryonic development and in protecting the body from damaged or ag...
