چکیده

Objective: The aim of this study was to determine the changes and underlying mechanisms of erectile organ structure and function in hyperlipidemic rats. Materials and Methods: Sixteen male rats were fed a normal diet to serve as controls, and the other sixteen rats were induced to develop hyperlipidemia with a high fat diet. After 4 months, the penile tissues were harvested and assessed for smooth muscle content. Molecular markers of phenotypic modulation were examined by qRT-PCR and western blotting. Results: Serum total cholesterol, triglyceride, and low density lipoprotein cholesterol levels were significantly higher in hyperlipidemic rats than those in control rats (P<0.01 for all). However, the high density lipoprotein cholesterol level was significantly reduced (P<0.01). Erectile function was significantly lower in hyperlipidemic rats than in control rats. There was no significant difference in the content of smooth muscle cells between the hyperlipidemic group and the control group. The expression of CCSMC phenotypic markers, such as calponin, smooth muscle myosin heavy chain 11, and myocardin was markedly lower, whereas osteopontin protein expression was significantly higher in hyperlipidemic rats than in control rats. Conclusion: CCSMCs possesses the ability to modulate the phenotype under hyperlipidemic conditions, which could have a key role in the pathogenesis of erectile dysfunction.

Phenotypic modulation of rat corpus cavernosum smooth muscle cells induced by platelet-derived growth factor-BB

Background: Phenotypic modulation within corpus cavernosum smooth muscle cells (CCSMCs) plays a critical role in the pathogenesis of erectile dysfunction (ED), yet little is known regarding the effects of plateletderived growth factor-BB (PDGF-BB) on CCSMCs phenotype. Objective: The aim of the present study was to evaluate the effect of PDGF-BB on phenotypic modulation of CCSMCs. Method: Cell v...

Phenotypic Modulation of Corpus Cavernosum Smooth Muscle Cells in a Rat Model of Cavernous Neurectomy

BACKGROUND Patients undergoing radical prostatectomy (RP) are at high risk for erectile dysfunction (ED) due to potential cavernous nerve (CN) damage during surgery. Penile hypoxia after RP is thought to significantly contribute to ED pathogenesis. AIM We previously showed that corpora cavernosum smooth muscle cells (CCSMCs) undergo phenotypic modulation under hypoxic conditions in vitro. Her...

Organization and relative content of smooth muscle cells, collagen and elastic fibers in the corpus cavernosum of rat penis.

PURPOSE The corpus cavernosum smooth muscle and extracellular matrix are essential for normal penile erection and are implicated in erectile dysfunction. Although investigations of these issues have used the rat corpus cavernosum, organization of its components is to date not well known. We characterized and quantified the smooth muscle cells and the main extracellular matrix components of the ...

The Protective Effect of Salidroside on Hypoxia-Induced Corpus Cavernosum Smooth Muscle Cell Phenotypic Transformation

Salidroside, a major active ingredient isolated from Rhodiola rosea, has a long application in Chinese medical history. It has widely demonstrated effects on fatigue, psychological stress, and depression and exhibits potential antihypoxia activity. Emerging evidence shows that hypoxia is an important independent risk factor for erectile dysfunction (ED). The aim of this study was to clarify the...

Application of Outlier Robust Nonlinear Mixed Effect Estimation in Examining the Effect of Phenylephrine in Rat Corpus Cavernosum

Ignoring two main characteristics of the concentration-response data, correlation between observations and presence of outliers, may lead to misleading results. Therefore the special method should be considered. In this paper in to examine the effect of phenylephrine in rat Corpus cavernosum, outlier robust nonlinear mixed estimation is used. in this study, eight different doses of phenylephrin...
