چکیده

Any survey of Type-I1 restriction endonucleases requires a consideration of the various specificities that are the hallmark of these enzymes. The latest compilation of the restriction-enzyme list contains more than 200 enzymes, which exhibit more than 50 unique specificities (Roberts, 1980). Because these enzymes have demonstrated their greatest value as biochemical tools, classification of these enzymes into groups based on common themes inherent within their varied specificities provides an attractive scaffolding for this presentation. The unique specificities of the currently known restriction endonucleases are listed in Table 1 based on the size of their recognition sites.* These recognition sequences vary in length from four to six base pairs. An important addition to the tetranucleotide group of enzymes has been made with the recent discovery of enzyme RsaI (GTAC) by J. F. Gardner & S. Kaplan (unpublished work). With the description of this recognition site, eight of the 16 possible tetranucleotide palindromic combinations have now been discovered. Another restriction endonuclease of interest listed within the tetra-

Restriction and modification enzymes and their recognition sequences

Since the last compilation of restriction endonucleases (110), more than 30 Type II restriction endonucleases have been discovered, including some valuable new specificities. These include Acyl (GPuCGPyC), Ddel (CTNAG), Fnu4HI (GCNGC), Rsal (GTAC), SphI (GCAGTC), and XmalH (CGGCCG). In addition, a number of new isoschizomers have been discovered and further information about the recognition seq...

Digestion of DNA with restriction endonucleases.

Restriction endonucleases recognize short DNA sequences and cleave double-stranded DNA at specific sites within or adjacent to the recognition sequences. Restriction endonuclease cleavage of DNA into discrete fragments is one of the most basic procedures in molecular biology. The first method presented in this unit is the cleavage of a single DNA sample with a single restriction endonuclease. A...

Identification of type II restriction and modification systems in Helicobacter pylori reveals their substantial diversity among strains.

A total of 22 type II restriction endonucleases with 18 distinct specificities have been identified in six Helicobacter pylori strains. Among these 18 specificities are three completely new endonucleases, Hpy178III, Hpy99I, and Hpy188I, that specifically cleave DNA at TCNNGA, CGWCG, and TCNGA sites, respectively. The set of endonucleases identified in each strain varies, but all have four- or f...

Restriction endonucleases from Herpetosiphon giganteus: an example of the evolution of DNA recognition specificity?

We describe the partial purification and characterisation of five Type II restriction endonucleases from two strains of Herpetosiphon giganteus. One of the activities, HgiJII, was the first enzyme found that cleaves DNA at the family of related sequences 5'-G-R-G-C-Y/C-3'. This enzyme may be related to the enzyme HgiAI from a different strain of the same species, and which cleaves at the sites ...

Cleavage of mispaired heteroduplex DNA substrates by numerous restriction enzymes.

The utility of restriction endonucleases as a tool in molecular biology is in large part due to the high degree of specificity with which they cleave well-characterized DNA recognition sequences. The specificity of restriction endonucleases is not absolute, yet many commonly used assays of biological phenomena and contemporary molecular biology techniques rely on the premise that restriction en...
