New mannose - specific lectins from garlic ( AZZium sativum ) and ramsons ( AZ & m ursinum ) bulbs *

چکیده
Two new mannose-binding lectins were isolated from garlic (Allium satiuum, ASA) and ramsons (All&m ursinum, AUA) bulbs, of the family Alliaceae, by affinity chromatography on immobilized mannose. The carbohydrate-binding specificity of these two lectins was studied by quantitative precipitation and hapten-inhibition assay. ASA reacted strongly with a synthetic linear (1 --$ 3)a-o-mannan and S. cerevisiae mannan, weakly with a synthetic (1 + 6)-o-D-mannan, and failed to precipitate with galactomannans from T. gropengiesseri and T. lactis-condensi, a linear mannopentaose, and murine IgM. On the other hand, AUA gave a strong reaction of precipitation with murine IgM, and good reactions with S. cerevisiae mannan and both synthetic linear mannans, suggesting that the two lectins have somewhat different binding specificities for n-o-mannosyl units. Of the saccharides tested as inhibitors of precipitation, those with (Y-(1 + 3)-linked mannosyl units were the best inhibitors of ASA, the a-(1 j 2)-, o-(1 * 4), and o-(1 + 6)-linked mannobioses and biosides having less than one eighth the affinity of the o-(1 + 3)linked compounds. The N-terminal amino acid sequence of ASA exhibits 79% homology with that of AUA, and moderately high homology (53%) with that of snowdrop bulb lectin, also an a-o-mannosyl-binding lectin.

A Comparative Study of Mannose-binding Lectins from the Amaryllidaceae and Alliaceae
A comparative study of the lectins from the families Amaryllidaceae and Alliaceae reveals many common features: all bind D-mannose exclusively and have similar molecular structures and amino acid compositions. All these lectins contain subunits of M, 11500-14000 which are not linked by disulphide bonds and occur as dimers (in Allium sativum, A. vineale, A. ursinum, A. moly, Narcissus pseudonarc...
متن کاملGarlic (Allium sativum) lectins bind to high mannose oligosaccharide chains.
Two mannose-binding lectins, Allium sativum agglutinin (ASA) I (25 kDa) and ASAIII (48 kDa), from garlic bulbs have been purified by affinity chromatography followed by gel filtration. The subunit structures of these lectins are different, but they display similar sugar specificities. Both ASAI and ASAIII are made up of 12.5- and 11.5-kDa subunits. In addition, a complex (136 kDa) comprising a ...
متن کاملThe Chemical Compositions of the Volatile Oils of Garlic (Allium sativum) and Wild Garlic (Allium vineale)
Garlic, Alliumsativum, is broadly used around the world for its numerous culinary and medicinal uses. Wild garlic, Allium vineale, has been used as a substitute for garlic, both in food as well as in herbal medicine. The present study investigated the chemical compositions of A. sativum and A. vineale essential oils. The essential oils from the bulbs of A. sativum, cultivated in Spain, were obt...
متن کاملCloning and Molecular Characterization of LECASAI Lectin Gene from Garlic (Allium Sativum L.)
Lectins have always demonstrated their role in bearing insecticidal activity. They are although considered to be toxic for mammals but some studies have reported that the presence of mannose binding region is believed to impart non-toxic nature to lectins against mammals including humans. In the present investigation, the research was conducted to elucidate the relationship of lectin gene LECAS...
