Recombinant Human Interferon-? Inhibits Adenovirus Multiplication

ثبت نشده
چکیده

The susceptibility of adenovirus to the inhibitory effect of human interferons in vitro was investigated. We tested recombinant human interferons-c~2, -/~1 and -7 against adenovirus serotypes 1 and 5 (group C), 3 and 7a (group B), and a wild strain isolated from an acutely ill child who later died. Pretreatment of WISH cells with interferon-? for 24 h induced a dose-dependent inhibition of multiplication of all adenovirus strains tested, the TCIDs0 varying from 25 to 90 IU/ml. Human interferon-c~ 2 was unable to decrease adenovirus multiplication, while interferon-/~l at 2000 IU/ml slightly lowered the yield of adenovirus. Similar results were obtained in HEp-2 and FS-4 cells, while A549 and peripheral blood mononuclear cells were insensitive to interferon-?. The difference between the effects of interferon-? and interferon-c~ and -,8 on adenovirus multiplication in vitro suggests that its mechanism of antiviral action is different.

Recombinant human interferon-gamma inhibits adenovirus multiplication without modifying viral penetration.

We have recently reported that adenovirus replication is inhibited by human recombinant interferon-gamma, but not by recombinant interferon-alpha, in a dose-dependent manner. The aim of this study was to determine whether the antiviral effect of recombinant interferon-gamma could be linked to interferon-induced alteration at the membrane level, inhibiting either adenovirus penetration of or rel...

Production of Recombinant Adenovirus Containing Human Interlukin-4 Gene

Objective(s) Recombinant adenoviruses are currently used for a variety of purposes, including in vitro gene transfer, in vivo vaccination, and gene therapy. Ability to infect many cell types, high efficiency in gene transfer, entering both dividing and non dividing cells, and growing to high titers make this virus a good choice for using in various experiments. In the present experiment, a reco...

Involvement of tumor necrosis factor alpha in intracellular multiplication of Legionella pneumophila in human monocytes.

We investigated the role of tumor necrosis factor alpha (TNF-alpha) in human peripheral monocytes infected with Legionella pneumophila in vitro. Exogenous TNF-alpha significantly inhibited the intracellular multiplication of the bacterium. This effect was concentration and time dependent and was abrogated by anti-TNF antibodies. TNF-alpha levels in the culture supernatants were low but were enh...

Overexpression of Recombinant Human Beta Interferon (rhINF-β) in Periplasmic Space of Escherichia coli

Human Interferon β (INF-β) is a member of cytokines family which different studies have shown its immunomodulatory and antiviral activities. In this study an expression vector was designed and constructed for expression of human INF-β-1b either in shake flasks and bench top bioreactor. The designed vector was constructed based upon pET-25b(+) with T7 promoter. Recombinant human beta interferon ...
