چکیده

Stable ferro-forms of hemoglobins containing various hemes such as dimethyl proto-, dhnethyl meso-, etio-, meso-, hemato-, and deuteroheme were prepared by combination of human globin with hemes, followed by reduction with dithionite and purification with carboxymethyl Sephadex column. Sedimentation coefficients of these hemoglobins were ah near to 4.0 S and chemical analysis showed that 4 moles of heme were bound per mole of hemoglobin. Esteritlcation of propionyl carboxyl groups of heme did not change the absorption spectra of hemoglobin; absorption spectra of dimethyl protohemoglobin were essentially identical with those of protohemoglobin, and absorption spectra of dimethyl mesohemoglobin with those of mesohemoglobin. Absorption spectra of etiohemoglobin, in which the propionic acid groups of mesoheme were substituted by ethyl groups, were similar to those of mesohemoglobin. Parameters of oxygen equilibrium, namely oxygen affinity, n in Hill’s equation, and Bohr effect, of hemoglobin containing hemes which lack free carboxyl groups were the same as those for the corresponding hemoglobin with free propionic acid groups. Oxygen affinities of meso-, deutero-, hemato-, and protohemoglobin, in which heme side chains at positions 2 and 4 are ethyl, hydrogen, hydroxyethyl, and vinyl groups, respectively, were in the ratio of 5 :2 : 1.3 : 1, corresponding to the reverse order of the negative inductive effects of the groups at positions 2 and 4. These results suggest the effect of P electron of a porphyrin ring on the oxygen binding.

Exchange of heme among hemoglobins and between hemoglobin and albumin.

Intact heme groups undergo exchange between molecules of human hemoglobin under physiological conditions. This phenomenon requires the presence of ferrihemoglobin and is blocked by heme ligands or the prior binding of hemoglobin to haptoglobin. The kinetics of heme exchange between hemoglobins A and F allows a calculation of the rate constants for the dissociation of heme and globin. The rate o...

A human hemoglobin with lowered oxygen affinity and impaired heme-heme interactions.

The reactivity of the iron in the four hemes of the mammalian hemoglobin molecule depends largely on the interactions between the prosthetic groups and the amino acid chains of the globin (1, 2). Normal configuration of heme groups in the abnormal human hemoglobins, therefore, does not preclude anomalies of their oxygen affinity which might arise from aberrations in the globin. In most of the m...

The role of hemoglobin heme loss in Heinz body formation: studies with a partially heme-deficient hemoglobin and with genetically unstable hemoglobins.

A number of mutant hemoglobins are inordinately unstable, denaturing in circulating red cells into Heinz bodies, resulting in congenital Heinz body hemolytic anemia (CHBHA). We have emphasized that most such hemoglobins involve amino acid substitutions at sites neighboring the heme group of the beta-polypeptide chain, and have shown that heme binding to globin is diminished thereby. Thus, hemes...

The binding of N-ethylmaleimide by human hemoglobin and its effect upon the oxygen equilibrium.

The reaction of human and horse hemoglobin with mercurials leads to considerable changes in the oxygen equilibrium of the proteins (2, 3). Two important features of the normal equilibrium can be altered by mercurials and silver (4). These are (a) the facilitating effect of oxygenation of one heme on the oxygenation of certain other hemes, and (b) the discharge of protons associated with oxygena...
