Microsatellite Polymorphisms in the Ovine Prion Protein Gene in Relation to Prp Genotypes

چکیده
The aims of the study were to detect the polymorphisms of three microsatellite sites: S11, S15, and S24 in the ovine PRNP and to asses their relationship with prion protein (PrP) genotypes in three sheep breeds. To identify 15 PrP genotypes based on polymorphisms at codons 136, 154, and 171, PCR-RFLP analyses were applied. The microsatellite sites were amplified. For each microsatellite two or three alleles per site in homoand heterozygotic combinations were observed. In the group of scrapie resistant sheep (NSP1), the genotype 152/152 bp of the S11 microsatellite, 179/179 bp of S15, and 144/144 bp of S24 occurred with the highest frequencies. The results of Fisher’s exact test showed that associations between PrP genotypes and microsatellite alleles and genotypes in the PRNP were statistically significant (P<0.001). In conclusion, the investigated microsatellite sites may be used as additional genetic markers in prion protein gene variability studies in sheep.

Breeding for resistance to scrapie: Detection of polymorphisms in the prion protein gene in German sheep breeds
Scrapie is a fatal neurodegenerative disease of sheep and goats belonging to the group of transmissible spongiform encephalopathies (TSE). TSEs are characterised by distinctive pathology, the involvement of a hostencoded prion protein (PrP) in disease development and genetic control of susceptibility. Polymorphisms in three different codons of the ovine prion protein (PrP) gene (136, 154, 171) ...
متن کاملIdentification of a novel ovine PrP polymorphism and scrapie-resistant genotypes for St. Croix White and a related composite breed.
Susceptibility to scrapie is primarily controlled by polymorphisms in the ovine prion protein gene (PRNP). Here, we report a novel ovine exon three PRNP polymorphism (SNP G346C; P116), its association with the ovine ARQ allele (P116A136R154Q171), and two new genotypes (PARQ/ARR; PARQ/ARQ) for the St. Croix White (SCW) breed and a related composite (CMP) breed developed for meat production. The ...

BACKGROUND The cellular prion protein PrP(C) is encoded by the Prnp gene. This protein is expressed in the central nervous system (CNS) and serves as a precursor to the misfolded PrP(Sc) isoform in prion diseases. The prototype prion disease is scrapie in sheep, and whereas Prnp exhibits common missense polymorphisms for V136A, R154H and Q171R in ovine populations, genetic variation in mouse Pr...

Bovine spongiform encephalopathy (BSE) is a fatal infectious neurodegenerative disease in cattle, characterized by the accumulation of an abnormal, proteaseresistant prion protein (PrPSc) in the brain. BSE is similar to scrapie in sheep and goats and Creuzfeldt-Jakob disease in humans. Susceptibility in cattle hasbeen shown to be under the influence of two polymorphic locations, which are...
متن کاملPrion protein (PrP) gene polymorphisms and breeding for resistance to scrapie in Polish Merino sheep
Genetic susceptibility or resistance of sheep to scrapie is associated with polymorphisms particularly in the three codons 136, 154 and 171 of the ovine prion protein (PrP) gene. The aim of the study was to reveal the PrP haplotype and genotype distribution in the Polish Merino breed and to make a forecast of the application of a scrapie resistance breeding programme for this breed. PCR-RFLP an...
