چکیده

The increasing energy demand and the anticipated decline in crude oil production has led to an immense search for new energy sources. Plant cell walls contain lignocellulose that conserve great amounts of energy. These polysaccharides are of high importance for the search of renewable energy sources. Pretreatment of the cell wall is necessary in order to hydrolyse it to its component sugars. Once degraded to monomeric sugars it can be fermented to either ethanol or biogas through established fermentation technologies. The aim of this thesis was to compare and evaluate some of the methods used for sacchrification of lignocellulose. Three treatments where compared to determine which is highest yielding. These are enzymatic hydrolysis, microwave irradiation and steam explosion. Wheat straw was used as substrate and hydrolysed by three commercial enzyme mixtures. Samples were pretreated before the enzymatic reaction with either microwave or steam explosion. Results showed that a treatment of either microwave irradiation or steam explosion combined with enzyme hydrolysis gives the highest yield in monomeric sugars. The conclusions that can be drawn are that mechanical pretreatment increases yield drastically but is insufficient in its self. Further enzymatic treatment of wheat straw is necessary to obtain high amounts of simple sugars. sockerarter från lignocellulosa.

Efficient whole-cell-catalyzing cellulose saccharification using engineered Clostridium thermocellum

BACKGROUND Cost-efficient saccharification is one of the main bottlenecks for industrial lignocellulose conversion. Clostridium thermocellum naturally degrades lignocellulose efficiently using the cellulosome, a multiprotein supermolecular complex, and thus can be potentially used as a low-cost catalyst for lignocellulose saccharification. The industrial use of C. thermocellum is restrained due...

Lignocellulosic saccharification by a newly isolated bacterium, Ruminiclostridium thermocellum M3 and cellular cellulase activities for high ratio of glucose to cellobiose

BACKGROUND Lignocellulosic biomass is one of earth's most abundant resources, and it has great potential for biofuel production because it is renewable and has carbon-neutral characteristics. Lignocellulose is mainly composed of carbohydrate polymers (cellulose and hemicellulose), which contain approximately 75 % fermentable sugars for biofuel fermentation. However, saccharification by cellulas...

Overcoming Biomass Recalcitrance by Combining Genetically Modified Switchgrass and Cellulose Solvent-Based Lignocellulose Pretreatment

Decreasing lignin content of plant biomass by genetic engineering is believed to mitigate biomass recalcitrance and improve saccharification efficiency of plant biomass. In this study, we compared two different pretreatment methods (i.e., dilute acid and cellulose solvent) on transgenic plant biomass samples having different lignin contents and investigated biomass saccharification efficiency. ...

In muro deacetylation of xylan affects lignin properties and improves saccharification of aspen wood

BACKGROUND Lignocellulose from fast growing hardwood species is a preferred source of polysaccharides for advanced biofuels and "green" chemicals. However, the extensive acetylation of hardwood xylan hinders lignocellulose saccharification by obstructing enzymatic xylan hydrolysis and causing inhibitory acetic acid concentrations during microbial sugar fermentation. To optimize lignocellulose f...
