چکیده

In eukaryotes, members of the phosphoinositide-3-kinase-related protein kinase (PIKK) family co-ordinate the cellular response to DNA damage. But how do these important kinases detect DNA damage and relay this information to the DNA repair and checkpoint machinery?

Analysis of ionizing radiation-induced DNA damage and repair in three-dimensional human skin model system.

Knowledge of cellular responses in tissue microenvironment is crucial for the accurate prediction of human health risks following chronic or acute exposure to ionizing radiation (IR). With this objective, we investigated the radio responses for the first time in three-dimensional (3D) artificial human skin tissue microenvironment after gamma-rays radiation. IR-induced DNA damage/repair response...

TopBP1 activates ATR through ATRIP and a PIKK regulatory domain.

The ATR (ATM and Rad3-related) kinase and its regulatory partner ATRIP (ATR-interacting protein) coordinate checkpoint responses to DNA damage and replication stress. TopBP1 functions as a general activator of ATR. However, the mechanism by which TopBP1 activates ATR is unknown. Here, we show that ATRIP contains a TopBP1-interacting region that is necessary for the association of TopBP1 and ATR...

Valosin-Containing Protein Phosphorylation at Ser in Response to DNA Damage

The response of eukaryotic cells to DNA damage includes the activation of phosphatidylinositol-3 kinase–related kinases (PIKK), such as ATM, ATR, and DNA-dependent protein kinase (DNA-PK). These three kinases have very similar substrate specificities in vitro , but in vivo , their substrates overlap only partially. Several in vivo substrates of ATM and ATR have been identified and almost all of...

Phosphatidylinositide 3-kinase (PI3K) and PI3K-related kinase (PIKK) activity contributes to radioresistance in thyroid carcinomas

Anaplastic (ATC) and certain follicular thyroid-carcinomas (FTCs) are radioresistant. The Phosphatidylinositide 3-kinase (PI3K) pathway is commonly hyperactivated in thyroid-carcinomas. PI3K can modify the PI3K-related kinases (PIKKs) in response to radiation: How PIKKs interact with PI3K and contribute to radioresistance in thyroid-carcinomas is unknown. Further uncertainties exist in how thes...
