چکیده

Mathematical modelling of metabolic pathway provides us a wide variety of information about behaviour of the system. The kinetic approach to the modelling of the systems is sometimes hampered by the fact that kinetic properties are imperfectly know that makes the structural approach more attractive. In order to make kinetic analysis of a metabolic pathway, construction of mathematical model describing its kinetics is a major part of the work. In the framework of metabolic kinetic theory, it is assumed that the rate of changes in the concentration i x of a metabolite i X is the sum of the r reaction rates, each weighted by corresponding stoichiometric coefficient of i X . Using v and x to denote the rate vector and concentration vector respectively, mathematical model for kinetics of a system can be written as dx Nv dt = where N is stoichiometric matrix which represents how the metabolites involved in the system combine. Derivation of conservation relationship which mainly depends on decomposition of stoichiometric matrix N plays important roles in constructing mathematical model of the system.

Metabolic engineering of lactic acid bacteria, the combined approach: kinetic modelling, metabolic control and experimental analysis.

Everyone who has ever tried to radically change metabolic fluxes knows that it is often harder to determine which enzymes have to be modified than it is to actually implement these changes. In the more traditional genetic engineering approaches 'bottle-necks' are pinpointed using qualitative, intuitive approaches, but the alleviation of suspected 'rate-limiting' steps has not often been success...

The evolution of control and distribution of adaptive mutations in a metabolic pathway.

In an attempt to understand whether it should be expected that some genes tend to be used disproportionately often by natural selection, we investigated two related phenomena: the evolution of flux control among enzymes in a metabolic pathway and properties of adaptive substitutions in pathway enzymes. These two phenomena are related by the principle that adaptive substitutions should occur mor...

The Effects of Aerobic Training on Serum Level of Liver Enzymes and Metabolic Syndrome Risk Factors in Young Inactive Women

Abstract        Background and Objectives: Nonalcoholic fatty liver disease is the most common chronic liver disorder and is also currently considered as the hepatic manifestation of metabolic syndrome. Regular exercise training may decrease fatty liver disease complications, although its impact is not yet clear. This study aimed to evaluate the effects of six...

Towards kinetic modeling of genome-scale metabolic networks without sacrificing stoichiometric, thermodynamic and physiological constraints.

Mathematical modeling is an essential tool for the comprehensive understanding of cell metabolism and its interactions with the environmental and process conditions. Recent developments in the construction and analysis of stoichiometric models made it possible to define limits on steady-state metabolic behavior using flux balance analysis. However, detailed information on enzyme kinetics and en...

The living cell is a complex system of interacting processes. The properties of the agents that facilitate these processes, such as enzymes, transporters and receptors, must be tuned to each other if the system is to behave harmoniously. The present chapter describes how the regulatory design of cellular subsystems that makes this harmonious behaviour possible can be visualized on a graph that ...
