چکیده

Metallothioneins are low-molecular-weight proteins capable of covalently binding heavy metal ions due to the presence of many cysteine residues in their sequences. We analyzed the predicted amino acid sequences of 19 metallothionein (7 from Arabidopsis thaliana and 12 from Oryza sativa) and their promoter sequences in silico in order to determine the potential regulatory cis-elements present in the promoters of metallothionein genes, from which it is possible to determine the putative functions of these genes. The PlantCARE and PLACE databases provided information about the putative regulatory elements in the metallothionein promoters. Metal response element sequences were found in the promoters of eleven O. sativa and two Arabidopsis metallothionein genes. Copper response elements were identified in both model plants, usually in many copies, particularly in O. sativa. Both the high cysteine content and the presence of metal response motifs in the promoters support the suggestion that metallothioneins play a key role in metal detoxification. The most common putative element in the analyzed promoters was CIRCADIAN, which was present in five A. thaliana and eight O. sativa sequences. The methyl jasmonate response sequence, root-specific expression element and drought response element were found only in O. sativa metallothioneins. Light and low temperature response elements, biotic and abiotic stress elements, an abscisic acid-responsive element and an ethylene-responsive element occur in selected metallothionein promoters of both species. A few promoters have putative organand cell-specific regulatory elements. The presence of many different motifs in the promoters of the Arabidopsis and O. sativa genes implies that metallothioneins are general stress response proteins with many important functions in plants, including regulation of their normal development and adaptation to changing environmental conditions.

Arabidopsis leaf plasma membrane proteome using a gel free method: Focus on receptor–like kinases

The hydrophobic proteins of plant plasma membrane still remain largely unknown.  For example in the Arabidopsis genome, receptor-like kinases (RLKs) are plasma membrane proteins, functioning as the primary receptors in the signaling of stress conditions, hormones and the presence of pathogens form a diverse family of over 610 genes. A limited number of these proteins have appeard in pr...

O-30: Comparing Expression Patterns of Endometrial Genes in Implantation Failures and Recurrent Miscarriages with Fertile Couples Following ICSI/IVF Using in Silico Analysis

Background: To screen and diagnose patients with recurrent abortions and implantation failure after IVF/ICSI, differentially expressed genes of endometrium through DNA microarrays were monitored. Materials and Methods: Microarray expression profile of GSE26787 dataset from GEO database was used to analyze gene expression profiles of 15 endometrial biopsy samples- five from control fertile (CF) ...

AKT family and miRNAs expression in IL-2-induced CD4+T cells

Objective(s): Study of non-coding RNAs is considerable to elucidate principal biological questions or design new therapeutic strategies. miRNAs are a group of non-coding RNAs that their functions in PI3K/AKT signaling and apoptosis pathways after T cell activation is not entirely clear. Herein, miRNAs expression and their putative targets in the mentioned pathways were studied in the activated ...

In silico screening of G-Quadruplex Structures in Wilms tumor 1 Gene Promoter

Introduction: X-ray diffraction studies have revealed that guanines in a DNA stands may be arranged in quartet and form a structure called G-quadruplexs. Bioinformatics studies suggested the formation of G-quadruplex structure in human crucial genes, including Wilms tumor 1 (WT1). The aim of this study was to in silico analysis of the guanine-rich sequence in the promoter region of the WT1 gene...

Systematic Analysis and Identification of Stress-Responsive Genes of the NAC Gene Family in Brachypodium distachyon

Plant-specific NAC proteins are one of the largest families of transcription factors in plants, and members of this family have been characterized with roles in the regulation of diverse biological processes, including development and stress responses. In the present study, we identified 101 putative NAC domain-encoding genes (BdNACs) through systematic sequence analysis in Brachypodium distach...
