Reaction of the sulfhydryl group with an oxidation product of beta-3,4-dihydroxyphenyl-L-alanine.

چکیده
The sulfhydryl group plays a role in the biological formation of melanin. Mechanisms suggested for the inverse relationship between the sulfhydryl group and the formation of melanin in mammalian skin include inhibition of tyrosinase and reaction with precursors of melanin (1, 2). However, knowledge in this field is fragmentary, and definitive proof of mechanism is lacking (3). The present work is a study in vitro of the reaction between an oxidation product of p-3,4-dihydroxyphenyl-L-alanine and the sulfhydryl group of cysteine or glutathione in the presence of tyrosinase at pH 7.4.

متن کاملKinetic and inhibition studies on catechol-O-methyltransferase affinity labelling by N-(3,4-dihydroxyphenyl)maleimide.
Initial velocity and product inhibition studies have been performed on soluble catechol-O-methyltransferase which has been partially purified from pig liver. The results are consistent with an ordered reaction mechanism, in which S-adenosyl-L-methionine (AdoMet) is the leading substrate. The enzyme is irreversibly inhibited by maleimide derivatives in a biphasic manner, which suggests a differe...
متن کاملA Crystallization-Induced Asymmetric Transformation using Racemic Phenyl Alanine Methyl Ester Derivatives as Versatile Precursors to Prepare Amino Acids
L-Tyrosine and L-Dopa are the precursors in the biological synthesis of amine neurotransmitters. On the other hand, phenylalanine as an aromatic amino acid (AAA) is a precursor in the synthesis of L-Tyrosine and L-Dopa. For some substrates such as amino acids, resolution by the formation of diastereomers offers an attractive alternative. Among different methods in this case, crystallization-ind...
متن کاملModelling of L-DOPA Oxidation Catalyzed by Laccase
Enzymatic oxidation of 3,4-dihydroxyphenyl-L-alanine (L-DOPA) with laccase from Trametes versicolor was investigated. The highest enzyme activity at pH 5.4 and at 25 °C was found. The reaction kinetics and the effect of dissolved oxygen concentration on the reaction rate were evaluated. A mathematical model, comprised of double-substrate Michaelis-Menten kinetics and mass balances for L-DOPA an...
متن کاملSynthesis, characterization and selective oxidation using a new copper (II) Schiff base complex derived from Alanine and 4-chloro3- formyl coumarin
A novel Schiff-base ligand (L: 2-[(4-chloro-2-oxo-2H-chromen-3-ylmethylene)-amino] propionic acid) was prepared from the reaction of 4-chloro3-formylcoumarin and alanine amino acid. Copper (II) complex was synthesized from the reaction of the ligand with Cu (OAc)2. H2O in ethanol. The ligand and its metal complex were characterized by elemental analysis (CHN), ICP, thermal analysis (TGA), Fouri...
