Significant manipulative procedures to reduce immunogenicity of human islet allografts.

چکیده
Immunofluorescence studies and mixed Iymphocyte islet culture (MLlC) tests were performed using cryopreserved crude human islet preparations l and various HLAas well as cell type-specific monoclonal antibodies in order to (a) locate the potentially immunogenic cells and (b) demonstrate their functional relevance before and after antibody incubation. Microscopic evaluation of exocrine and endocrine tissue components of crude islet preparations was greatly facilitated by the islet-specific dithizone staining technique.

Mixed lymphocyte islet culture (MLIC) and its use in manipulation of human islet alloimmunogenicity.
Human islets have been transplanted clinically to reverse diabetes, though with poor overall success (reviewed in Hering, Bretzel and Federlin 1988). Inferring from experimental islet transplantation, it is presently suggested that methods for separating the endocrine from the exocrine pancreas portion and/or serological tissue manipulations prior to islet transplantation should be sought in or...
متن کاملMHC-mismatched islet allografts are vulnerable to autoimmune recognition in vivo.
When transplanted into type 1a diabetic recipients, islet allografts are subject both to conventional allograft immunity and, presumably, to recurrent autoimmune (islet-specific) pathogenesis. Importantly, CD4 T cells play a central role both in islet allograft rejection and in autoimmune disease recurrence leading to the destruction of syngeneic islet transplants in diabetic NOD mice. However,...
متن کاملHuman Immune System Development and Rejection of Human Islet Allografts in Spontaneously Diabetic NOD-Rag1 IL2r null Ins2 Mice
RESEARCH DESIGN AND METHODS—We backcrossed the Ins2 mutation onto the NOD-Rag1 IL2r null strain and determined 1) the spontaneous development of hyperglycemia, 2) the ability of human islets, mouse islets, and dissociated mouse islet cells to restore euglycemia, 3) the generation of a human immune system following engraftment of human hematopoietic stem cells, and 4) the ability of the humanize...
متن کاملCo-Transplantation of VEGF-Expressing Human Embryonic Stem Cell Derived Mesenchymal Stem Cells to Enhance Islet Revascularization in Diabetic Nude Mice
Background: Pancreatic islet transplantation has emerged as a promising treatment for type I diabetes. However, its efficacy is severely hampered due to poor islet engraftment and revascularization, which have been resulted to partially loss of transplanted islets. It has been shown that local delivery of vascular endothelial growth factor (VEGF) could accelerate transplanted islet revasculari...
متن کاملPermanent survival of fully MHC-mismatched islet allografts by targeting a single chemokine receptor pathway.
Chemokine receptor blockade can diminish the recruitment of host effector cells and prolong allograft survival, but little is known of the role of chemokine receptors in promoting host sensitization. We engrafted fully allogeneic islets into streptozotocin-treated normal mice or mice with the autosomal recessive paucity of lymph node T cell (plt) mutation; the latter lack secondary lymphoid exp...
