Presence of basic fibroblast growth factor in the early Xenopus embryo

چکیده

Mesoderm-inducing activity can be extracted from Xenopus embryos, eggs or whole ovary. It binds to heparin and can be neutralized by heparin or anti-bFGF but not by anti-TGF/?. Two molecular forms can be identified by Western blotting and have molecular weights of about 19 and 14K. The content in embryos is about 7 units g~' (approximately 7 ng ml~*) which would be sufficient for it to be acting as an endogenous inducer of ventral mesoderm. Attempts to detect TGF/5-like inducing factors in embryos were not successful.

Molecular characterization of Xenopus embryo heparan sulfate. Differential structural requirements for the specific binding to basic fibroblast growth factor and follistatin.

Enzymatic elimination of heparan sulfate (HS) causes abnormal mesodermal and neural formation in Xenopus embryos, and HS plays an indispensable role in establishing the embryogenesis and tissue morphogenesis during early Xenopus development (Furuya, S., Sera, M., Tohno-oka, R., Sugahara, K., Shiokawa, K., and Hirabayashi, Y. (1995) Dev. Growth Differ. 37, 337-346). In this study, HS was purifie...

cDNA cloning and developmental expression of fibroblast growth factor receptors from Xenopus laevis.

The heparin-binding growth factors constitute a family of homologous polypeptides including basic and acidic fibroblast growth factors (FGFs). These factors participate in a variety of processes, including wound healing, angiogenesis, neuronal survival, and inductive events in the early amphibian embryo. We have isolated three closely related species of cDNA clones for Xenopus FGF receptors. On...

Novel HOX, POU and FKH genes expressed during bFGF-induced mesodermal differentiation in Xenopus.

Cells from the cap of the animal hemisphere of the early Xenopus embryo are determined to form ectodermal lineages. When these cells are explanted and cultured in the presence of various growth factors a change in fate to cells of mesodermal lineage can be observed. Proteins of the fibroblast growth factor (FGF) family belong to this class of fate altering compounds. The ability of FGFs to chan...

Molecular cloning of Xenopus HGF cDNA and its expression studies in Xenopus early embryogenesis

We isolated Xenopus HGF cDNA and examined its expression pattern in Xenopus early embryos and their dissected parts. Xenopus HGF consists of 710 amino acids and contains four kringle domains and serine protease-like structure just like mammalian HGF. Northern blot analysis showed that expression of Xenopus HGF mRNA starts at the late gastrula stage and its level increases during the period of l...

Localized and inducible expression of Xenopus-posterior (Xpo), a novel gene active in early frog embryos, encoding a protein with a 'CCHC' finger domain.

Xenopus-posterior (Xpo) is a gene that is activated at or shortly after the midblastula transition (MBT). The RNA accumulates to a relatively low level, which remains constant until gastrulation, then rapidly and transiently increases in posterior ectoderm and mesoderm. A single copy of a putative finger motif, of the 'CCHC' type, is located near the carboxyl terminus. One or two copies of simi...
