چکیده

A series of novel zoledronic acid (ZL) derivatives 1-hydroxy-3-(2-methyl-1H-imidazol-1-yl)propane-1,1-diyldiphosphonic acid (MIPrDP), 1-hydroxy-4-(2-methyl-1H-imidazol-1-yl)butane-1,1-diyldiphosphonic acid (MIBDP), and 1-hydroxy-5-(2-methyl-1H-imidazol-1-yl)pentane-1,1-diyldiphosphonic acid (MIPeDP) were prepared and successfully labeled with Tc in high labeling yields. The in vitro stability and in vivo biodistribution of Tc-MIPrDP, Tc-MIBDP and Tc-MIPeDP were investigated and compared. The biodistribution studies indicate that the radiotracer Tc-MIPrDP has highly selective uptake in the skeletal system and rapid clearance from soft tissues. The present findings indicate that Tc-MIPrDP holds great potential for use in bone imaging.

Advances in Drug Design of Radiometal-Based Imaging Agents for Bone Disorders

Nuclear medicine bone imaging has been the optimum diagnosis for the detection of bone disorders because the lesion could be detectable before the appearance of symptomatic and radiographic changes. Over the past three decades, (99m)Tc-MDP and (99m)Tc-HMDP have been used as bone scintigraphic agents because of their superior biodistribution characteristics, although they are far from optimal fr...

Comparison of state-of-the-art atlas-based bone segmentation approaches from brain MR images for MR-only radiation planning and PET/MR attenuation correction

Introduction: Magnetic Resonance (MR) imaging has emerged as a valuable tool in radiation treatment (RT) planning as well as Positron Emission Tomography (PET) imaging owing to its superior soft-tissue contrast. Due to the fact that there is no direct transformation from voxel intensity in MR images into electron density, itchr('39')s crucial to generate a pseudo-CT (Computed Tomography) image ...

Historical perspectives on the clinical development of bisphosphonates in the treatment of bone diseases.

Bisphosphonates (formerly termed diphosphonates) were first synthesized in the late 1800s; however, their clinical use has been relatively recent. The bisphosphonates' affinity for hydroxyapatite crystal surface led Procter and Gamble to test these compounds in dental, then medical applications. With key input from university researchers, this led to the medical use of the first bisphosphonate,...

The use of bisphosphonates in the management of bone involvement from solid tumours and haematological malignancies – a European survey

Bone metastases in patients with solid tumours (ST) and bone lesions in patients with haematological malignancies (HM) are common. Associated skeletal-related events (SREs) cause severe pain, reduced quality of life and place a burden on health care resources. Bone-targeted agents can reduce the risk of SREs. We evaluated the management of bone metastasis/lesions in five European countries (Fra...

The Beneficial Effects of Bisphosphonate-enoxacin on Cortical Bone Mass and Strength in Ovariectomized Rats

Osteoporosis is a major age-related bone disease characterized by low bone mineral density and a high risk of fractures. Bisphosphonates are considered as effective agents treating osteoporosis. However, long-term use of bisphosphonates is associated with some serious side effects, which limits the widespread clinical use of bisphosphonates. Here, we demonstrate a novel type of bone-targeting a...
