چکیده

26 Antimicrobial susceptibility testing (AST) of clinical isolates of Nocardia is recommended to 27 detect resistance to commonly used antimicrobial agents; such testing is complicated by 28 difficulties in inoculum preparation and test interpretation. In this study, six laboratories 29 performed repetitive broth microdilution testing on single strains of Nocardia brasiliensis, 30 Nocardia cyriacigeorgica, Nocardia farcinica, Nocardia nova and Nocardia wallacei. For each 31 isolate, a total of 30 microdilution panels from three different lots were tested at most sites. The 32 goal of the study was to determine the interand intra-laboratory reproducibility of susceptibility 33 testing of this group of isolates. Acceptable agreement [> 90% agreement at (±) 1 dilution of the 34 minimum inhibitory concentration (MIC) mode] was found for amikacin, ciprofloxacin, 35 clarithromycin and moxifloxacin. After eliminating MIC values from single laboratories whose 36 results showed the greatest deviation from those of the remaining laboratories, acceptable 37 agreement was also found for amoxicillin/clavulanic acid, linezolid, minocycline and 38 tobramycin. Results showed unsatisfactory reproducibility of broth microdilution testing of 39 ceftriaxone with N. cyriacigeorgica and N. wallacei, tigecycline with N. brasiliensis and N. 40 cyriacigeorgica and sulfonamides with N. farcinica and N. wallacei. N. nova ATCC BAA-2227 41 is proposed as a quality control organism for AST of Nocardia sp., and the use of a disk 42 diffusion test for sulfisoxazole is proposed as a check of the adequacy of the inoculum and to 43 confirm sulfonamide MIC results. 44 on N ovem er 3, 2017 by gest ht://jcm .sm .rg/ D ow nladed fom

Multisite reproducibility of Etest for susceptibility testing of Mycobacterium abscessus, Mycobacterium chelonae, and Mycobacterium fortuitum.

A multicenter study was conducted to assess the inter- and intralaboratory reproducibility of the Etest for susceptibility testing of the rapidly growing mycobacteria. The accuracy also was evaluated by comparing Etest results to those obtained by broth microdilution. Ten isolates (four of the Mycobacterium fortuitum group, three of Mycobacterium abscessus, and three of Mycobacterium chelonae) ...

Multisite reproducibility of results obtained by the broth microdilution method for susceptibility testing of Mycobacterium abscessus, Mycobacterium chelonae, and Mycobacterium fortuitum.

A multicenter study was conducted to assess the interlaboratory reproducibility of broth microdilution testing of the more common rapidly growing pathogenic mycobacteria. Ten isolates (four Mycobacterium fortuitum group, three Mycobacterium abscessus, and three Mycobacterium chelonae isolates) were tested against amikacin, cefoxitin, ciprofloxacin, clarithromycin, doxycycline, imipenem, sulfame...

Multisite reproducibility of colorimetric broth microdilution method for antifungal susceptibility testing of yeast isolates.

MICs of fluconazole and amphotericin B were determined independently for 100 coded yeast isolates by each of six laboratories to determine reproducibility of results by using a colorimetric oxidation-reduction-based broth microdilution test. In addition, each site tested five quality control isolates on at least four different occasions during the study. Results agreed within a three-dilution r...

In vitro activities of linezolid against multiple Nocardia species.

Linezolid was tested by broth microdilution against 140 clinical Nocardia isolates belonging to seven species. The MIC at which 50% of the strains are inhibited (MIC50) and MIC90 for all species other than Nocardia farcinica were 2 and 4 microg/ml. Linezolid is the first antimicrobial agent demonstrated to be active against all Nocardia species.

Quality control and reference guidelines for CLSI broth microdilution susceptibility method (M 38-A document) for amphotericin B, itraconazole, posaconazole, and voriconazole.

Although standard conditions are available for testing the susceptibilities of filamentous fungi to antifungal agents by the Clinical and Laboratory Standards Institute (CLSI; formerly National Committee for Clinical Laboratory Standards) broth microdilution assay, quality control (QC) MIC limits have not been established for any mold-agent combination. This multicenter (eight-center) study doc...
