Regulatory regions of androgen-responsive genes.

چکیده
Androgens are presumed to influence gene transcription by modifying the association of the androgen receptor with specific nuclear sites. However, the details of this process are still obscure. Elucidation of the mechanisms involved is complicated by the diversity of androgen binding within targetcell nuclei and by a lack of understanding of the interaction o f androgen receptors with androgen-responsive genes, and is hampered by the unavailability as yet of androgen receptor protein purified to homogeneity. This report describes our endeavours in this area, with special reference to our studies on androgen mechanisms in rat ventral prostate and the interaction of androgen-receptor complexes with regions of the prostate binding protein (PBP) C3( 1) gene (see Parker et

Upstream Regulatory Elements, Potential Targets and Expression Patterns of Three Drought Responsive miRNAs in Two Grapevine Cultivars
MicroRNAs (miRNAs), as a group of non-coding small RNAs, play key roles in regulating the growth, development and response of plants to various stresses. In this study, the expression patterns of three drought responsive miRNAs (miR159c, miR160a,b and miR169v) were compared in both drought tolerant (Yaghuti) and drought sensitive (Bidanesefid) grapevine cultivars using qRT-PCR under drought str...
متن کاملA hierarchical network of transcription factors governs androgen receptor-dependent prostate cancer growth.
Androgen receptor (AR) is a ligand-dependent transcription factor that plays a key role in prostate cancer. Little is known about the nature of AR cis-regulatory sites in the human genome. We have mapped the AR binding regions on two chromosomes in human prostate cancer cells by combining chromatin immunoprecipitation (ChIP) with tiled oligonucleotide microarrays. We find that the majority of A...

The AR (androgen receptor) is known to influence the expression of its target genes by binding to different sets of AREs (androgen-response elements) in the DNA. One set consists of the classical steroid-response elements which are partial palindromic repeats of the 5'-TGTTCT-3' steroid-receptor monomer-binding element. The second set contains motifs that are AR-specific and that are proposed t...

The androgen receptor (AR) signaling pathway is a common therapeutic target for prostate cancer, because it is critical for the survival of both hormone-responsive and castrate-resistant tumor cells. Most of the detailed understanding that we have of AR transcriptional activation has been gained by studying classical target genes. For more than two decades, Kallikrein 3 (KLK3) (prostate-specifi...
متن کاملCell- and gene-specific regulation of primary target genes by the androgen receptor.
The androgen receptor (AR) mediates the physiologic and pathophysiologic effects of androgens including sexual differentiation, prostate development, and cancer progression by binding to genomic androgen response elements (AREs), which influence transcription of AR target genes. The composition and context of AREs differ between genes, thus enabling AR to confer multiple regulatory functions wi...
