Lipid Screening in Single Microalgae Using Hydrogel Microcapsule Arrays
نویسندگان
چکیده
We demonstrated a microwell array-based microfluidic system, allowing in situ analysis of the lipid content of individual microalgal cells in alginate hydrogel microcapsules. Alginate hydrogel microcapsules with a mean diameter of 26 μm, which encapsulate a single microalga stained with nondestructive lipophilic green fluorescent dye, BODIPY 505/515, were successfully docked inside the microwell. By direct observation of the BODIPY fluorescence response from a single live alga in each microwell, stochastic heterogeneity in the lipid content among the same species was verified in a physiological condition with high viability. The proposed microcapsule-based platform for screening of lipid-rich microalgae has great potential to expand research fields of environmental engineering.

Microalgae, a group of microorganisms that grow using sunlight as the sole energy source and carbon dioxide as an only carbon source, have been considered as a feedstock of choice for the production of biofuels such as biodiesel. To explore the economic feasibility of such application, however, many technical hurdles must first be overcome; the selection and/or screening of competent species ar...

Microcapsule arrays attract a lot of interest due to their potential applications in sensing technology. A strategy for fabricating diverse microcapsule arrays through covalent linking is reported here. The self-assembly of microcapsules was directed by using a poly(allylamine hydrochloride) (PAH)-patterned template, which was created via microcontact printing. The microcapsules with PAH as the...

The involvement of algal chemical cues in the pre-ingestive selection of food particles in Crassostrea gigas was studied using a new approach. Live cells of two microalgal species, Nitzschia closterium and Tetraselmis suesica, were separately entrapped in small alginate microcapsules using an emulsification/internal gelation method. Microcapsule size was adjusted to be within the range of parti...
متن کاملA Single-Step Method for Rapid Extraction of Total Lipids from Green Microalgae
Microalgae produce a wide range of lipid compounds of potential commercial interest. Total lipid extraction performed by conventional extraction methods, relying on the chloroform-methanol solvent system are too laborious and time consuming for screening large numbers of samples. In this study, three previous extraction methods devised by Folch et al. (1957), Bligh and Dyer (1959) and Selstam a...

By virtue of the biocompatibility and physical properties of hydrogel, picoliter-sized hydrogel microcapsules have been considered to be a biometric signature containing several features similar to that of encapsulated single cells, including phenotype, viability, and intracellular content. To maximize the experimental potential of encapsulating cells in hydrogel microcapsules, a method that en...
