چکیده

The in vitro sensitivity to chloroquine and pyrimethamine of 19 culture-adapted southern African reference isolates of Plasmodium falciparum was determined using a 48-hour assay. Four isolates collected in KwaZulu, Natal, were sensitive to chloroquine, and one of these was sensitive to the drug in vivo. Eight isolates from KwaZulu or Mozambique were resistant to chloroquine in vitro. Six of these isolates were chloroquine-resistant in varying degrees in vivo. Four of five isolates from north-eastern Transvaal and two clinically chloroquine-resistant Malawian isolates were resistant to chloroquine in vitro. A wide range of pyrimethamine susceptibilities was detected (0.01 mumol/l to greater than 3.0 mumol/l), although most isolates were inhibited at 0.1 mumol/l, indicating a low level of resistance. These results confirm the presence of both chloroquine and pyrimethamine resistance in the endemic areas of South Africa. This has serious implications for the prophylaxis and treatment of P. falciparum malaria in South Africa.

Treatment of Malaria Parasitaemia in Infants and their Mothers

Malaria is an infection sustained by three parasites namely: Plasmodium falciparum, Plasmodium vivax, and Plasmodium ovale. Plasmodium falciparum is the most common and virulent parasite. These parasites are present in different areas of the sub-Saharan African countries and Asia. In 2010, there were an estimated 219 million cases of malaria resulting in 660,000 deaths and, approximately, two-t...

Resistance of ten Thai isolates of Plasmodium falciparum to chloroquine and pyrimethamine by in vitro tests.

In vitro drug resistance tests of ten isolates of Plasmodium falciparum from three different collection points in Central Thailand have been carried out, and the results compared with those of similar tests with a drug-sensitive West African isolate. Judged by concentration of drug tolerated, the Thai isolates appeared to be about 10 times as resistant to chloroquine, and usually about 10(5) ti...

Reemergence of chloroquine-sensitive Plasmodium falciparum malaria after cessation of chloroquine use in Malawi.

In 1993, Malawi became the first African country to replace chloroquine with sulfadoxine-pyrimethamine nationwide in response to high rates of chloroquine-resistant falciparum malaria. To determine whether withdrawal of chloroquine can lead to the reemergence of chloroquine sensitivity, the prevalence of the pfcrt 76T molecular marker for chloroquine-resistant Plasmodium falciparum malaria was ...

Monitoring the drug-sensitivity of Plasmodium falciparum in coastal towns in Madagascar by use of in vitro chemosensitivity and mutation detection tests.

The dissemination of mutant and resistant strains of Plasmodium falciparum makes a considerable contribution to the spread of drug-resistant malaria. Populations around harbours and airports could be particularly exposed to Plasmodium isolates introduced with imported cases of malaria. The use of chloroquine as well as the use of and sulfadoxine/pyrimethamine is currently an effective method fo...

In vitro activity of dihydroartemisinin against clinical isolates of Plasmodium falciparum in Yaounde, Cameroon.

The in vitro activities of dihydroartemisinin (the biologically active metabolite of artemisinin derivatives), chloroquine, monodesethylamodiaquine (the biologically active metabolite of amodiaquine), quinine, mefloquine, halofantrine, and pyrimethamine were assessed in 65 African isolates of Plasmodium falciparum from Yaounde, Cameroon using an isotopic microtest. The 50% inhibitory concentrat...
