چکیده
This study was focused on purification and characterization of keratinase from Bacillus thuringiensis TS2, isolated from feather dumping soil site. Zymogram of crude enzyme on native-PAGE presented bands with keratinase activity of molecular weights is 41 kDa. In purification steps 2.69 fold purification was achieved after 80% ammonium precipitation of the with 34.42% recovery. In further purification steps a 5.69 fold purified keratinase was recovered by Sephadex G-75 chromatography with 21.23% of recovery. The specific activity of purified enzyme was 970.15.54 U/mg. Kinetic studies indicated that substrate specificity of TS2 keratinase was towards various natural and synthetic proteolytic substrates but inactive against collagen and keratin. These findings suggested that the keratinase may have much industrial application.

متن کاملCharacterization of the keratinolytic activity of indigenous Bacillus subtilis keratinase
Keratins are water insoluble proteins of our environment. Being extremely resistant to degradation by proteolytic enzymes, keratins are digested mainly by alkali and keratinase enzymes. The aim of present study was to characterize the indigenous keratinase for potentials of keratin-degrading activities. We report the purification and characterization of keratinase from Bacillus subtilis isolate...
متن کاملNovel keratinase from Bacillus subtilis S14 exhibiting remarkable dehairing capabilities.
We report the isolation of a keratinolytic-producing Bacillus subtilis strain and the characterization of the exceptional dehairing properties of its subtilisin-like keratinase. This enzyme can be an alternative to sodium sulfide, the major pollutant from tanneries, and may completely replace it. Its unique nonactivity upon collagen enhances its industrial potential.

Bacillus thuringiensis (Bt) has been extensively used in biopesticidal formulations due to its safe environmental and human health records. The widespread use of Bt is often challenged by production as well as formulation costs which are in direct contact with downstream processing, i.e. the separation/purification step. Downstream separation/purification efficacy governs the marketabi...

This study reports the purification and biochemical characterization of thermostable and acidic-pH-stable α-amylase from Bacillus sp. Iranian S1 isolated from the desert soil (Gandom-e-Beryan in Lut desert, Iran). Amylase production was found to be growth associated. Maximum enzyme production was in exponential phase with activity 2.93 U ml-1 at 50°C and pH 5. The enzyme was purified by isoprop...
