چکیده

Corticotropin-releasing factor (CRF) and urocortin (UCN) are potent regulators of Purkinje cell dendritic development including spines and synapses. Here, we demonstrate that CRF and UCN increase dendritic spine density in cerebellar Purkinje cells. Specific spine types in Purkinje cells are selectively affected: CRF promotes the mushroom type of spines, whereas at the same time UCN the thin type. Also, CRF and UCN are involved in the regulation of activity-dependent refinement of synaptic connections of Purkinje cells. Both peptides lead to increasing the length of the active zone and the postsynaptic density (PSD) in synapses between Purkinje cells and parallel fiber terminals. Finally, CRF and UCN are involved in processing the topological matching of presynaptic and postsynaptic specializations of synapses. Slices treated with CRF and UCN show less mismatched synapses in contrast to control slices.

The postnatal developmental expression pattern of urocortin in the rat olivocerebellar system.

Urocortin belongs to the family of corticotropin-releasing factor (CRF)-like peptides, which play an important role in sensorimotor coordination. CRF induces locomotor activity, and urocortin has an inhibitory effect. Here, we document the regional and subcellular localization of urocortin in the developing rat cerebellum to compare it with CRF. During the first postnatal week, urocortin immuno...

Effects of Cadmium on Cellular and Subcellular Structures of Cerebellar Purkinje Cells in Four-Day Postnatal Developing Rat

Purpose: This experiment was conducted to evaluate the effects of Cd on cellular and subcellular aspects of cerebellar Purkinje cells in developing rats. Materials and methods: forty adult female Wistar rats served as subjects in this experiment. The animals were assigned randomly to four groups: control I, control II, experiment I and experiment II. The experiment group I and II were injected...

Stereological Estimation of Granule Cell Number and Purkinje Cell Volume in the Cerebellum of Noise-Exposed Young Rat

In spite of the existing reports on behavioural and biochemical changes related to the cerebellum due to noise stress, not much is known about the effect of noise stress on the neuronal changes in the cerebellum. The present study aims at investigating the effects from one week noise exposure on granule cell number and Purkinje cell volume within the neonate rat cerebellum.15-day-old male Wista...

P 18: Alterations of Electrophysiological Activity of Cerebellar Pukinje Cells of Rats Under Harmaline Toxicity

Introduction: Beta-carboline alkaloids of P. harmala are shown to have immune-modulatory effects in several studies. Extracts of this plant have significant anti-inflammatory effect via the inhibition of some inflammatory mediators including PGE2 and TNF-α. In postmortem studies, structural alterations to the cerebellum have been recognized, including Purkinje cell loss being re...

Corticotropin-releasing Factor (CRF) and Urocortin Promote the Survival of Cultured Cerebellar GABAergic Neurons Through the Type 1 CRF Receptor

Corticotropin releasing factor (CRF) is known to be involved in the stress response and in some degenerative brain disorders. In addition, CRF has a role as a neuromodulator in adult cerebellar circuits. Data from developmental studies suggest a putative role for CRF as a trophic factor during cerebellar development. In this study, we investigated the trophic role for CRF family of peptides by ...
