The stoicheiometric relationships between electron transport, proton translocation and adenosine triphosphate synthesis and hydrolysis in mitochondria.

چکیده
The validity of the conclusion that the mitochondrial redoxand ATP-driven proton pumps translocate 2 H+ ions per ‘high-energy phosphate bond’ has been challenged on theoretical and experimental grounds. This review attempts to highlight the difficulties of the present position and to introduce an alternative model within the framework of the chemiosmotic hypothesis based on a stoicheiometry of 3 H+ (or possibly 4 H+) per ‘high-energy phosphate bond’.

Stoichiometry of adenosine triphosphate-driven proton translocation in bovine heart submitochondrial particles.
Inward-directed proton translocation driven by ATP hydrolysis was investigated in bovine heart submitochondrial particles under conditions where no net pH change occurred upon ATP hydrolysis. A biphasic time course of ATP hydrolysis, due to the presence of adenylate kinase activity, was observed by following the release of 32Pi from [P,r32P]ATP under comparable conditions. An equation was deriv...

A transmembrane electrochemical proton gradient is widely held to constitute the link between electron transport and ATP synthesis in biological membranes (Boyer et al., 1977), but much debate surrounds the issue of the stoicheiometry of H+ movement that either generates (by electron transport) or dissipates (by ATP synthesis) this proton gradient (proton-motive force, Ap) (Brand, 1977). Our es...

ATP hydrolysis and synthesis by the F(0)F(1)-ATP synthase are coupled to proton translocation across the membrane in the presence of magnesium. Calcium is known, however, to disrupt this coupling in the photosynthetic enzyme in a unique way: it does not support ATP synthesis, and CaATP hydrolysis is decoupled from any proton translocation, but the membrane does not become leaky to protons. Unde...
متن کاملRespiratory Complex I in Bos taurus and Paracoccus denitrificans Pumps Four Protons across the Membrane for Every NADH Oxidized*
Respiratory complex I couples electron transfer between NADH and ubiquinone to proton translocation across an energy-transducing membrane to support the proton-motive force that drives ATP synthesis. The proton-pumping stoichiometry of complex I (i.e. the number of protons pumped for each two electrons transferred) underpins all mechanistic proposals. However, it remains controversial and has n...
متن کاملThe respiratory complex I of bacteria, archaea and eukarya and its module common with membrane-bound multisubunit hydrogenases.
The proton-pumping NADH:ubiquinone oxidoreductase, also called complex I, is the first of the respiratory complexes providing the proton motive force which is essential for energy consuming processes like the synthesis of ATP. Homologues of this complex exist in bacteria, archaea, in mitochondria of eukaryotes and in chloroplasts of plants. The bacterial and mitochondrial complexes function as ...
