چکیده

F R O M the results of experiments involving autoradiography of stretched H3 thymidine-labelled salivary gland chromosomes of Drosophila melanogaster, and a consideration of previously published evidence, STEFFENSEN (1963) has concluded that DNA (deoxyribonucleic acid) is absent from the interband regions of these chromosomes. In opposition to this conclusion are the results presented in this report of examinations in the fluorescence microscope of acridine orange stained, ribonuclease digested salivary glands of Drosophila melanogaster, which provide direct evidence for the presence of DNA in interband regions of polytene chromosomes.

Identical Functional Organization of Nonpolytene and Polytene Chromosomes in Drosophila melanogaster

Salivary gland polytene chromosomes demonstrate banding pattern, genetic meaning of which is an enigma for decades. Till now it is not known how to mark the band/interband borders on physical map of DNA and structures of polytene chromosomes are not characterized in molecular and genetic terms. It is not known either similar banding pattern exists in chromosomes of regular diploid mitotically d...

On the Presence of Dna in Larval Salivary Gland Nucleoli in Drosophila Melanogaster

In the course of recent experiments on Drosophila salivary gland chromosomes with both fluorescent dyes and isotopic labeling, we have found evidence which bears on the question of nucleolar DNA. We would like to describe these observations in support of the growing number of reports on the presence of DNA in nucleoli (1-5; see reference 4 for additional references) and in view of

Visualization of unconstrained negative supercoils of DNA on polytene chromosomes of Drosophila.

Bulk DNA within the eukaryotic genome is torsionarily relaxed. However, unconstrained negative supercoils of DNA have been detected in few local domains of the genome through preferential binding of psoralen. To make a genome-wide survey for such domains, we introduced biotinylated psoralen into Drosophila salivary glands and visualized it on polytene chromosomes with fluorescent streptavidin. ...

Localization of ecdysterone on polytene chromosomes of Drosophila melanogaster.

Ecdysterone has been crosslinked in situ to polytene chromosomes of salivary glands of Drosophila melanogaster by photoactivation. The crosslinked hormone has been localized on the chromosomes by indirect immunofluorescence microscopy. At different developmental stages the hormone was detected at different chromosomal loci. These chromosomal sites correspond to ecdysterone-inducible puff sites....

The Drosophila salivary gland chromocenter contains highly polytenized subdomains of mitotic heterochromatin.

Peri-centromeric regions of Drosophila melanogaster chromosomes appear heterochromatic in mitotic cells and become greatly underrepresented in giant polytene chromosomes, where they aggregate into a central mass called the chromocenter. We used P elements inserted at sites dispersed throughout much of the mitotic heterochromatin to analyze the fate of 31 individual sites during polytenization. ...
