چکیده

A rapid, simple, and sensitive method for quantitation of ethchlorvynol by gas Chromatography has been developed. instrument response is linear to a concentration of 200 mg/liter (2.0-mi sample of serum). Day-to-day (20 days) precision of a 15 mg/liter serum pool was 0.85 mg/liter (SD) and 5.6% (CV). For 30 samples analyzed by both the gas chromatographic method and a colorimetric method, the correlation plot had a slope of 0.989, x and y intercepts near zero, and a correlation coefficient of 0.997. Mean recovery of ethchlorvynol added to serum was 87%. Subsequently, all standards were prepared in serum, to compensate for the nonquantitative yield and to help assure accuracy. Sera from healthy controls and from hospitalized patients contain no endogenous interfering substances, nor did any of a series of commonly prescribed drugs interfere.

Ethchlorvynol (Placidyl) intoxication and its treatment by hemodialysis.

Twenty hours after ingesting 15-25 g. of ethchlorvynol, a 37-year-old woman was admitted comatose and in shock. The blood level of ethchlorvynol was 21.6 mg. % (method of Wallace). Supportive measures were instituted and hemodialysis, carried out for 10 hours, removed 5.49 g. of the drug. The post-dialysis blood level was 9.05 mg. % and the rate of dialysance was 50.5 ml./min. Only 0.6 g. of th...

Simple and sensitive high performance liquid chromatographic method for the simultaneous quantitation of the phenylalanine in human plasma

Phenylalanine (Phe) is the most reliable indicator for the diagnosis of phenylketonuria (PKU). The purpose of this study is to establish a reliable and quick method for the assignment of Phe in peripheral capillary blood from newborns and children by high performance liquid chromatography with ultraviolet detection (HPLC-UV). PKU is an inborn error of metabolism characterized by the inability o...

Gas-chromatographic determination of disopyramide and its mono-N-dealkylated metabolite in serum with use of a nitrogen-selective detector.

I describe a rapid, simple routine gas-liquid chromatographic method for simultaneous isothermal quantitation of disopyramide and its mono-N-dealkylated metabolite. The method involves one extraction step with dichloromethane, derivatization of the metabolite, and gas chromatography with an OV-17 liquid phase. Standard curves were linear over the concentration range tested (0.1-25 mg/L) and the...

Letter: Ethchlorvynol interference with oxidimetric determination of ethanol in urine.

Gas-chromatographic methods for ethanol quantitation provide greater specificity than traditional oxidimetric methods. The specificity of oxidimetric methods such as those of Kozelka-Hine (1) and Widmark (2) rely on the arguments that: (a) the sample came from a living person and (b) drugs or chemicals that would appreciably react with potassium dichromate could not be expected to be present in...

Rapid method for quantitative determination of propoxyphene in serum by gas-liquid chromatography.

Rapid, accurate, and precise gas-chromatographic methods are reported for measurement of propoxyphene (“Darvon”) in serum. A sample of 5 ml of serum is required for quantitation in blood after a therapeutic dose of 130-195 mg of propoxyphene; in cases of overdose of propoxyphene, only 1 ml of serum is required in a “toxic method” variation. Neither serum from healthy controls or from hospitaliz...
