چکیده

The incidence of invasive fungal infections has increased significantly in the last several years, which can be explained by the widely using of wide-spectrum antibiotics and immunosuppressant, invasive procedures and medical implant devices. Among the fungal pathogens of humans, Candida represented by Candida albicans, is one of the species that are the most frequently associated with biofilm infection, particularly which on medical implants, and this has a significant impact on morbidity and mortalit [1]. invasive procedures and medical implant devices C. albicans which can cause both superficial and systemic infections, is one of the main pathogenic microorganisms caused nosocomial infection. Data in 2002 from the US National Nosocomial Infections Surveillance System ranked C. albicans as the fourth most common cause of bloodstream infection, behind coagulaseIn vivo Models for Candida Albicans Biofilms Study Wenrui Gu1, Dongmei Xu2, Dongmei Guo3, Liuping Zhang4 and Shujuan Sun5* 1School of Pharmaceutical Sciences, Shandong University, Jinan, 250012, Shandong Province, People’s Republic of China 2Department of Nephrology, Qianfoshan Hospital Affiliated to Shandong University, Jinan, 250014, P.R. In vivo efficacy of anidulafungin against mature Candida albicans biofilms in a novel rat model of catheter-associated Candidiasis.

The present study demonstrates the efficacy of anidulafungin on mature Candida albicans biofilms in vivo. One hundred fifty-seven catheter fragments challenged with C. albicans were implanted subcutaneously in rats. After formation of biofilms, rats were treated with daily intraperitoneal injections of anidulafungin for 7 days. Catheters retrieved from treated animals showed reduced cell number...

Gallic acid/hydroxypropyl-β-cyclodextrin complex: Improving solubility for application on in vitro/ in vivo Candida albicans biofilms

The aim of this study was to increase the solubility of gallic acid (GA) for the treatment of Candida albicans biofilm, which is very difficult to treat and requires high drug concentrations. Cyclodextrins (CDs) were used for this purpose. Complexes were evaluated by phase-solubility studies, prepared by spray drying and characterized by drug loading, scanning electron microscopy (SEM) and diff...

Activities of systemically administered echinocandins against in vivo mature Candida albicans biofilms developed in a rat subcutaneous model.

This study addresses the effects of micafungin, caspofungin, and anidulafungin against Candida albicans biofilms developed in a subcutaneous catheter rat model system. Doses of 5, 10, and 30 mg/kg (of body weight)/day (the last only for micafungin) were given intravenously for 5, 7, and 10 days. All three echinocandins caused a significant reduction of the Candida cell numbers on the implanted ...

Towards non-invasive monitoring of pathogen–host interactions during Candida albicans biofilm formation using in vivo bioluminescence

Candida albicans is a major human fungal pathogen causing mucosal and deep tissue infections of which the majority is associated with biofilm formation on medical implants. Biofilms have a huge impact on public health, as fungal biofilms are highly resistant against most antimycotics. Animal models of biofilm formation are indispensable for improving our understanding of biofilm development ins...

Candida albicans biofilm development on medically-relevant foreign bodies in a mouse subcutaneous model followed by bioluminescence imaging.

Candida albicans biofilm development on biotic and/or abiotic surfaces represents a specific threat for hospitalized patients. So far, C. albicans biofilms have been studied predominantly in vitro but there is a crucial need for better understanding of this dynamic process under in vivo conditions. We developed an in vivo subcutaneous rat model to study C. albicans biofilm formation. In our mod...
