IL-15R expression on CD8 T cells is dispensable for T cell memory

چکیده
The generation and maintenance of immunological memory requires the activation, expansion, and persistent proliferation of antigen-specific T cells. Recent work suggests that IL-15 may be important for this process. Surprisingly, we now find that expression of the high-affinity receptor for IL-15, IL-15R , on T cells is dispensable for the generation or maintenance of memory CD8 T cells. By contrast, IL-15R expression on cells other than T cells is absolutely critical for this function. These findings may be related to IL-15R ’s ability to present IL-15 in trans to low-affinity IL15R c receptors on memory CD8 T cells. These unexpected results provide insights into how IL-15R supports memory CD8 T cells.

Distinct cell types control lymphoid subset development by means of IL-15 and IL-15 receptor alpha expression.
IL-15 and the IL-15 receptor (IL-15R)alpha chain are essential for normal development of naive CD8 T cells, intestinal intraepithelial lymphocytes (IEL), and natural killer (NK)/NK/T cells. However, whether IL-15R alpha expression by these subsets is necessary for their production and which cell type needs to produce IL-15 to drive development are unknown. We analyzed the requirements for IL-15...

Following influenza virus infection, memory CD8 T cells are found in both lymphoid and nonlymphoid organs, where they exhibit striking differences in survival. We have assessed persistence, phenotype, and function of memory CD8 T cells expressing the same TCR in the airways, lung parenchyma, and spleen following influenza virus infection in mice. In contrast to memory CD8 T cells in the spleen,...

IL-15 stimulates the proliferation of memory phenotype CD44(high)CD8(+) T cells and is thought to play a key role in regulating the turnover of these cells in vivo. We have investigated whether IL-15 also has the capacity to affect the life span of naive phenotype (CD44(low)) CD8(+) T cells. We report that IL-15 promotes the survival of both CD44(low) and CD44(high) CD8(+) T cells, doing so at ...
متن کاملThe IL-15 receptor chain cytoplasmic domain is critical for normal IL-15R function but is not required for trans-presentation
IL-15 is critical for natural killer (NK)–cell development and function and for memory CD8 T-cell homeostasis. The IL-15 receptor consists of IL-15R , IL-2R , and the common cytokine receptor chain ( c). IL-15R is known to “trans-present” IL-15 to an IL-2R / c heterodimeric receptor on responding cells to initiate signaling. To investigate the importance of the IL-15R cytoplasmic domain, we gen...
متن کاملNo requirement of trans presentations of IL-15 for human CD8 T cell proliferation.
The trans presentation of IL-15 by cells expressing the specific high-affinity receptor α-chain (IL-15Rα) to cells expressing the signaling receptor β-chain and γ-chain is essential for the generation and maintenance of CD8 memory T cells, NK cells, and NKT cells in an in vivo mouse system. We have also demonstrated in vitro that cell-surface IL-15Rα on cells expressing all the receptor compone...
