چکیده
BACKGROUND The rat model of radiation-induced carcinogenesis is widely used to estimate breast cancer risk, but it is unknown whether mammary terminal end buds (TEBs), the hypothesized target of chemical carcinogens, are also targets of radiation-induced carcinogenesis. MATERIALS AND METHODS Female Sprague-Dawley rats were irradiated with X-rays (2 Gy). The morphology, bromodeoxyuridine (BrdU)-labelling index and pyknotic index of mammary ductal termini were investigated from 6 hours to 8 weeks post-irradiation. RESULTS The irradiated rats showed a significantly smaller decrease in TEB numbers than the controls 4-8 weeks after irradiation. The BrdU-labelling and pyknotic indices initially decreased and increased, respectively, recovered to control levels by 3 days post-irradiation, and remained significantly higher until 8 weeks and 4 weeks post-irradiation, respectively. One case of ductal hyperplasia was observed at 8 weeks. CONCLUSION Irradiation resulted in a persistent proliferation of TEBs, which may be associated with neoplastic transformation via a temporal differentiation arrest.

Prenatal TCDD and predisposition to mammary cancer in the rat.
Prenatal exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) was investigated for its potential to predispose to breast cancer. Analysis of mammary gland differentiation and cell proliferation were used as biomarkers. Timed pregnant Sprague-Dawley CD rats were gavaged with 1 microg TCDD/kg on day 15 post-conception. Control animals were treated with the same volume of vehicle (sesame oil) on...
متن کاملEffect of tamoxifen on preneoplastic cell proliferation in N-nitroso-N-methylurea-induced mammary carcinogenesis.
N-Nitroso-N-methylurea (NMU) is an effective carcinogen for the induction of mammary carcinoma in the rat. Tamoxifen (TAM), used as a chemopreventive agent to reduce tumor incidence, has been well studied using this model. We have utilized the rat mammary carcinoma model to assess the effect of TAM on preneoplastic changes. Fifty-day-old virgin female Sprague-Dawley rats were randomized by weig...

متن کاملCell proliferation and tumor formation induced by eserine, an acetylcholinesterase inhibitor, in rat mammary gland.
Environmental chemicals may be involved in the etiology of breast cancer. There is substantial evidence that breast cancer risk is associated with prolonged exposure to female hormones. Among these hormonal influences a leading role is attributed to the ovarian hormone estradiol, since breast cancer does not develop in the absence of ovaries. The rat mammary gland has special characteristics th...
متن کاملInfluence of differentiation and cell kinetics on the susceptibility of the rat mammary gland to carcinogenesis.
The susceptibility of the rat mammary gland to carcinogenesis decreases with aging and is nullified by pregnancy and lactation. This work was carried out to determine whether these differences in susceptibility to carcinogenesis are the result of variations in cell kinetics induced by both aging and parity. Cell cycle (Tc) and growth fraction (GF) were studied in the mammary gland epithelium of...
