چکیده

Received January 16,1978 SUMMARY: Rat ovarian cells were exposed to cholera enterotoxin, and the effect on progesterone synthesis as well as on protein kinase stimulation was examined. Cholera enterotoxin stimulated ovarian steroidogenesis in a dose dependent manner similar to that of hCG. The stimulation of protein kinase by cholera toxin was followed by a lag period, whereas hCG effect was imnediate. Mixed gangliosides, when added to the incubation medium, blocked the cholera enterotoxin-stimulated protein kinase activity and abolished the decrease in exogenous [sH] cyclic AMP receptor activity brought about by the toxin. In contrast, under similar experimental conditions ganglioside addition elicited no effect on protein kinase activation produced by hCG or LH. The data suggest that gangliosides do not appear to be directly involved in gonadotropin binding to ovarian cell membrane and subsequent mediation of physiological response. The available evidence suggests that adenosine 3':5' cyclic monophosphate (cyclic AMP) may be the intracellular mediator of gonadotropin mediated steroidogenesis in ovarian cells (l-6). Previous studies from this laboratory have shown that gonadotropins stimulate protein kinase in rat ovarian cells (4, 5). To evaluate further on the role of protein kinase in gonadotropin stimulated steroidoqenesis, we have investigated the effects of cholera enterotoxin on protein kinase activation in rat ovarian cells. This agent was chosen because cholera enterotoxin has been shown to mimic the action of several peptide hormones in their target tissues (7, 8). Furthermore, the existence of sequence analogies between cholera enterotoxin, LH, hCG, FSH and TSH (9, 10) suggests that all these agents may act through similar mechanisms and possibly through the involvement of qangliosides. Recent studies on ganglioside inhibition of LH and hCG binding to testis plasma membrane further

Receptor-Mediated Gonadotropin Action in Ovary

The effect of gangliosides and cholera enterotoxin on 1251 choriogonadotropin binding, cAMP formation, protein kinase activation and progesterone production have been studied in isolated rat ovarian interstitial cells. A close correlation was observed between the amount of adenosine 3' : 5'monophosphate (CAMP) produced in the cells and the extent of the activation of protein kinase in response ...

Role of gangliosides in gonadotropin and cholera enterotoxin stimulated steroidogenesis in isolated rat ovarian cells.

F : Ovarian cells isolated from 26 day old rats responded to hCG IO rig/ml) and cholera enterotoxin (100 rig/ml) in vitro with a forty-five to -fifty-fold increase in progesterone production. Both cholera enterotoxin and hCG-stimulated progesterone response was accompanied by a lag period. The duration of the lag period in the production of the progesterone depended on the concentration ot gona...

Cholera toxin stimulates type II pneumocyte proliferation by a cyclic AMP-independent mechanism.

Cholera toxin (CT) stimulated DNA synthesis by low-density primary cultures of adult rat type II pneumocytes (T2P) in a dose-dependent manner, either in the presence or the absence of serum. In the presence of 1% rat serum, 1 microgram/ml CT also stimulated a 50% increase in cell number over 8 days of incubation (P<0.01); this was in addition to a 2-fold increase in cell number induced by the s...

Comparison of the action of Escherichia coli enterotoxin on the thymocyte adenylate cyclase-cyclic adenosine monophosphate system to that of cholera toxin and prostaglandin E1.

Mouse thymocytes were used to compare mechanisms by which Vibrio cholerae and heat-labile Escherichia coli enterotoxins activate the adenylate cyclase-cyclic adenosine monophosphate (AMP) system. Both enterotoxins had their time-delayed increase in cyclic AMP neutralized by antisera to V. cholerae or E. coli enterotoxin, blocked by low concentrations of ganglioside G(M1), and destroyed by prior...

Gonadotropin-controlled mammal oocyte meiotic resumption.

Fully grown mammalian oocytes resume meiosis as a consequence of rises in gonadotropin levels at the mid-cycle. The increase of cyclic adenosine 3',5'-monophosphate (cAMP) and the activation of protein kinase A (PKA), protein kinase C (PKC) and mitogen-activated protein kinase (MAPK) in cumulus cells are required for gonadotropins-induced meiotic resumption of oocytes. The various actions of cA...
