چکیده

FIGURE 1. Effect 0fGDPBS, CTP.,S, and GTP on 3H-mucin release. Confluent cultures (16 mm) which had been pretreated with streptolysin 0 were pulsed with H-glucosamine for 24 h and chased for 30 mm in the presence of the nucleotides (200 p.M GTP.,S, 200 iM GDP,,S, and 200 M GTP). In panel A, GDP,,S was added immediately before GTP S. Data represent means ± SE of4 culture dishes. Mucin release by GTP S was not significantly (p>0.05) different in the presence or absence of GDP S. Based on Stadent s t-test for unpaired samples.

Nucleotide-induced mucin release from primary hamster tracheal surface epithelial cells involves the P2u purinoceptor.

Mucin release by airway surface epithelial cells is regulated by extracellular adenosine triphosphate (ATP) via a P2 purinoceptor-mediated mechanism. The objective of the present experiment was to examine the possible involvement of uridine triphosphate (UTP) in this purinergic signal transduction pathway. Using primary hamster tracheal surface epithelial cells, ATP and UTP were compared in the...

Stimulation of mucin secretion from human bronchial epithelial cells by mast cell chymase.

AIM To investigate the effect of chymase on the mucin secretion from human bronchial epithelial cells. METHODS Primarily-cultured human bronchial epithelial (PCHBE) cells and normal human bronchial epithelial (NHBE) cells were cultured with chymase or other stimulus in a mixture of bronchial epithelial growth medium (BEGM) and Dulbecco's modified Eagle's medium (DMEM), and the quantities of s...

Dual oxidase 1-dependent MUC5AC mucin expression in cultured human airway epithelial cells.

Mucus hypersecretion is a prominent manifestation in patients with chronic inflammatory airway diseases. MUC5AC mucin is a major component of airway mucus, and its expression is modulated by a TNF-alpha-converting enzyme (TACE)-EGF receptor pathway that can be activated by reactive oxygen species (ROS). Dual oxidase 1 (Duox1), a homologue of glycoprotein p91(phox), is expressed in airway epithe...

Inhibition of mucin release from airway goblet cells by polycationic peptides.

In the present study, we investigated whether polycationic peptides affect mucin release from cultured airway goblet cells. Confluent primary hamster tracheal surface epithelial cells were metabolically radiolabeled with [3H]glucosamine for 24 h and chased for 30 min in the presence of varying concentrations of either poly-l-arginine (PLA) or poly-l-lysine (PLL) to assess the effects on [3H]muc...

Cell to Cell Communication in Response to Mechanical Stress via Bilateral Release of Atp and Utp in Polarized Epithelia

Airway epithelia are positioned at the interface between the body and the environment, and generate complex signaling responses to inhaled toxins and other stresses. Luminal mechanical stimulation of airway epithelial cells produces a propagating wave of elevated intracellular Ca(2+) that coordinates components of the integrated epithelial stress response. In polarized airway epithelia, this re...
