چکیده
By RALPH L. NACHMAN, HERBERT I. HoRowrrz AND RICHARD T. SILVER E SSENTIAL THROMBOCYTOSIS is a disorder of unknown etiology characterized by a marked increase in the number of circulating platelets, and max’ l)C related to the myeloproliferative diseases.1 In the course of this condition, many patients develop hemorrhagic and/or thrombotic complicalions, which may result from an abnormal number of platelets, from a qualitative defect in platelets or from t combination of both factors. If the platelets in this disorder are truly defective, the abnormality might be a consequence of deranged levels of certain intracellular constituents. In order to evaluate this possibility, the intraplatelet free amino acid composition was studied. We have also studied the pseudohyperkalemic phenomenon which represents a spurious elevation of serum potassium levels owing to potassium release from platelets when whole blood containing increased numbers of platelets clots in vitro. In addition, platelet factor 3 availability was evaluated in the thromboplastin generation test.

Thrombocytosis
Background Thrombocytosis is defined as an increase in the number of circulating platelets than the normal value of between 150,000 and 250,000 platelets/uL. The limit beyond which the state of platelets is usually marked in platelets is defined as 400,000/uL. Trombocytosis are divided into primary or autonomous, in the course of myeloproliferative disorders (essential thrombocythemia, polycyth...
متن کاملPatients with thrombocytosis have normal or slightly elevated thrombopoietin levels.
BACKGROUND AND OBJECTIVE The distinction between clonal and reactive thrombocytoses is a frequent problem and implies different therapeutic options. As thrombopoietin (TPO) is the main regulator of megakaryocytopoiesis and thrombopoiesis, we measured TPO levels in patients with thrombocytosis in an attempt to understand the regulation and potential utility of distinguishing thrombocytoses. DE...
متن کاملJak-2 mutation frequency in patients with thrombocytosis
Background: We aimed to investigate the etiologic causes and the existence of Janus kinase 2 mutation (JAK2) in cases with thrombocytosis. Methods: In this retrospective study, patients, who were admitted to hematology clinic with thrombocytosis between 2013 and 2015, were investigated in terms of the etiological causes of thrombocytosis and the existence of JAK2 mutation. Results: We retrospe...
متن کاملReticulated platelets are increased in chronic myeloproliferative disorders, pure erythrocytosis, reactive thrombocytosis and prior to hematopoietic reconstitution after intensive chemotherapy.
Reticulated platelets with a high RNA content represent the most recently released platelets and are regarded to reflect thrombopoiesis. In the present study we used flow cytometric analysis to determine the percentage of reticulated platelets in peripheral blood for patients with chronic myeloproliferative disorders (polycythemia vera, essential thrombocytosis) and acute myelogenous leukemia (...
متن کاملIncreased soluble and platelet-associated CD40 ligand in essential thrombocythemia and reactive thrombocytosis.
CD40 ligand (CD40L) is expressed on activated CD4(+) T lymphocytes and at the activated platelet surface. A circulating soluble form of CD40L (sCD40L) is generated by the way of a proteolytic cleavage. We measured sCD40L in the plasma of either healthy subjects; patients with inflammatory disorders and low, normal, or high platelet count (reactive thrombocytosis); or patients with essential thr...
