Asymmetric Subunit Organization of Heterodimeric Rous Sarcoma Virus Reverse Transcriptase ab: Localization of the Polymerase and RNase H Active Sites in the a Subunit

چکیده
The genes encoding the a (63-kDa) and b (95-kDa) subunits of Rous sarcoma virus (RSV) reverse transcriptase (RT) or the entire Pol polypeptide (99 kDa) were mutated in the conserved aspartic acid residue Asp 181 of the polymerase active site (YMDD) or in the conserved Asp 505 residue of the RNase H active site. We have analyzed heterodimeric recombinant RSV ab and aPol RTs within which one subunit was selectively mutated. When ab heterodimers contained the Asp 1813Asn mutation in their b subunits, about 42% of the wild-type polymerase activity was detected, whereas when the heterodimers contained the same mutation in their a subunits, only 7.5% of the wild-type polymerase activity was detected. Similar results were obtained when the conserved Asp 505 residue of the RNase H active site was mutated to Asn. RNase H activity was clearly detectable in ab heterodimers mutated in the b subunit but was lost when the mutation was present in the a subunit. In summary, our data imply that the polymerase and RNase H active sites are located in the a subunit of the heterodimeric RSV RT ab.

Studies on the reverse transcriptase of RNA tumor viruses. Structural relatedness of two subunits of avian RNA tumor viruses.
The structural relationship of the small (alpha) and large (beta) subunits of reverse transcriptase isolated from two avian RNA tumor viruses has been examined by tryptic peptide analysis. Comparison of the tryptic hydrolysates of the isolated subunits by two-dimensional separation on thin-layer cellulose plates indicates that (i) the alpha subunit of reverse transcriptase of avian myeloblastos...

A radioimmunoassay was developed that can detect and quantitate 3 ng or more of the avian RNA tumor virus reverse transcriptase. The assay detected no antigenic sites in Rous sarcoma virus alpha virions or in virions of a murine RNA tumor virus. About 70 molecules of reverse transcriptase were found per virion of avian myleloblastosis virus with this assay or with an assay based on antibody inh...
متن کاملIntrinsic properties of reverse transcriptase in reverse transcription. Associated RNase H is essentially regarded as an endonuclease.
The intrinsic properties of reverse transcriptase in reverse transcription were studied using a synthetic, partial ovalbumin mRNA with a synthetic DNA oligonucleotide annealed to the 3'-end of the RNA as a model substrate. With or without concomitant cDNA synthesis, the RNase H activity of avian myeloblastosis virus (AMV)-reverse transcriptase cleaved the substrate at a site which would leave a...
متن کاملStructural analysis of monomeric retroviral reverse transcriptase in complex with an RNA/DNA hybrid
A key step in proliferation of retroviruses is the conversion of their RNA genome to double-stranded DNA, a process catalysed by multifunctional reverse transcriptases (RTs). Dimeric and monomeric RTs have been described, the latter exemplified by the enzyme of Moloney murine leukaemia virus. However, structural information is lacking that describes the substrate binding mechanism for a monomer...
متن کاملSubunit-specific analysis of the human immunodeficiency virus type 1 reverse transcriptase in vivo.
The human immunodeficiency virus type 1 (HIV-1) reverse transcriptase (RT) is a heterodimer comprised of two structurally distinct subunits (p51 and p66). Since p51 and p66 are derived from the same coding region, subunit-specific structure-function studies of RT have been conducted exclusively by in vitro biochemical approaches. To study RT subunit function in the context of infectious virus, ...
