چکیده

Membrane-binding of a human blood clotting protein, factor VIII (FVIII), was studied by cryoelectron microscopy. Well visible gap junctions between adjacent phosphatydyl serine containing phospholipid vesicles were observed upon FVIII association. Digital images acquired with a JEOL2010FEG transmission electron microscope equipped with a 4x4k CCD camera were analysed with the Digital Micrograph software to study the lipid protein interactions and shed light onto the mechanism of FVIII membrane-binding. This study has been carried out in conjunction with ongoing structural electron microscopy studies of membrane-bound FVIII to better understand its role in blood coagulation and haemophilia.

Lack of recombinant factor VIII B-domain induces phospholipid vesicle aggregation: implications for the immunogenicity of factor VIII

Factor VIII (FVIII) is a multidomain blood plasma glycoprotein. Activated FVIII acts as a cofactor to the serine protease factor IXa within the membrane-bound tenase complex assembled on the activated platelet surface. Defect or deficiency in FVIII causes haemophilia A, a severe hereditary bleeding disorder. Intravenous administration of plasma-derived FVIII or recombinant FVIII concentrates re...

Helical organization of blood coagulation factor VIII on lipid nanotubes.

Cryo-electron microscopy (Cryo-EM)(1) is a powerful approach to investigate the functional structure of proteins and complexes in a hydrated state and membrane environment(2). Coagulation Factor VIII (FVIII)(3) is a multi-domain blood plasma glycoprotein. Defect or deficiency of FVIII is the cause for Hemophilia type A - a severe bleeding disorder. Upon proteolytic activation, FVIII binds to th...

Cryo-electron microscopy of coagulation Factor VIII bound to lipid nanotubes.

Factor VIII (FVIII) is a key protein in blood coagulation, deficiency or malfunction of which causes Haemophilia A. The sole cure for this condition is intravenous administration of FVIII, whose membrane-bound structure we have studied by Cryo-electron microscopy and image analysis. Self-assembled lipid nanotubes were optimised to bind FVIII at close to native conditions. The tubes diameter was...

Cryo-electron microscopy of extracellular vesicles in fresh plasma

INTRODUCTION Extracellular vesicles (EV) are phospholipid bilayer-enclosed vesicles recognized as new mediators in intercellular communication and potential biomarkers of disease. They are found in many body fluids and mainly studied in fractions isolated from blood plasma in view of their potential in medicine. Due to the limitations of available analytical methods, morphological information o...
