Embryotoxicity of Carbendazim in Rat and Hamster Micromass Cultures

The embryotoxic potential of the commonly used fungicide, methyl 2-benzimidazolecarbamate (MBC) was evaluated in rat and hamster embryo limb bud (LB) and midbrain (CNS) cell micromass cultures. Cultures were exposed to seven concentrations of 0.03, 0.12, 0.49, 1.95, 7.81, 31.25 and 125 μg MBC/ml. Developmental toxicity was assessed via the following parameters: cytotoxicity, by quantifying neutral red uptake; differentiation, by alcian blue staining of extracellular proteoglycans (LB) and by haematoxylin staining (CNS). Two endpoints 50% inhibition concentration for proliferation (IC50-P) and differentiation (IC50-D) were calculated. The mean IC50-D (μg/ml) were: in LB cultures 1.88 (rat) and 1.54 (hamster); in CNS cultures 0.66 (rat) and 0.90 (hamster). It means that both models can detect the same potential of dismorphogenic effects. Based on the results MBC was classified as strong embryotoxic compound.
