چکیده

A centrifugal method for ultrarapid freezing of cells and subcellular particles has been developed, in which small droplets are centrifuged at high speed through liquid nitrogen. When material such as water or blood was injected from a sYI·inge into a rotor, increased rotor speeds gave progressively smaller frozen droplets. Droplet size of materials introduced from a sprayer was uniform, smaller than the smallest droplets obtained from a syringe injection, and independent of rotor speed. Cell survival was found to be inversely related to increasing droplet size and directly related to rotor speed and warming rate.-Nat Cancer lnst Monogr 21: 415-430, 1966.

The Action of Extreme Cold on Leukemic Cells of Mice

Suspensions of leukemic cells of mice from three different strains of leukemia were subjected to rapid or slow freezing and rapid or slow thawing. Suspensions rapidly frozen to -196 degrees C. were in all cases innocuous, whereas those frozen slowly were capable of transmitting leukemia. The infectivity of slowly frozen material varied from an estimated 0.0001 per cent to 1 per cent of that of ...

Experimental verification of morphological instability in freezing aqueous colloidal suspensions.

We describe an experimental test of a new theory of the unidirectional freezing of aqueous colloidal suspensions. At low freezing speeds a planar ice lens completely rejects the particles, forming a steady-state compacted boundary layer in the liquid region. At higher speeds the planar interface becomes thermodynamically unstable and breaks down geometrically to trap bulk regions of colloid wit...

Reduction of mortality rates of Bacillus thuringiensis var. israelensis aqueous suspensions due to freezing and thawing.

When studying the behavior (carry, dispersion, persistence) of Bacillus thuringiensis var. israelensis (B.t.i.) formulations used in the treatment of rivers or streams for black fly control, a large number of samples containing small quantities of B.t.i. are required for proper analysis. Freezing is a useful procedure to prevent enzymatic alteration or bacterial growth in samples before bioassa...

Oxidative phosphorylation by mitochondrial suspensions after freezing and storage at low temperatures.

There is much evidence to suggest that the structural and functional integrity of many biochemical materials is unaltered by freezing if the rate of cooling is rapid and the storage temperature low (2-6). Isolated enzymes and mixtures of enzymes and their substrates, after rapid freezing at low temperatures, often retain their activities and in some cases show enhanced activity upon thawing (7-...

Mechanism of polymerization of the Salmonella O-antigen: utilization of lipid-linked intermediates.

Cell envelope fractions from Salmonella can utilize exogenous lipid-linked intermediates for the synthesis of polymeric O-antigen. We describe a method for preparing aqueous suspensions of lipid intermediates and show that freezing and thawing of cell envelope-lipid intermediate mixtures is required for efficient synthesis. The lipid intermediates move freely in the hydrophobic environment of t...
