Spectroscopic data for steroids subjected to Zak color reaction.

چکیده
HE COLOR TEST OF Zak et al. (1, 2, 3), in which a mixture of concentrated sulfuric acid, ferric chloride, and acetic acid is added to a solution of a steroid in glacial acetic acid, gives a purple color for steriods with the 5-ene,3$-ol grouping, and has proved useful for the clinical determination of cholesterol. These authors have qualitatively described the colors given by 22 other steroids in the visual spectral range. We have investigated the feasibility of qualitative and quantitative determinations in the range of 320 to 700 m of a number of steriods not previously studied by this reaction. In the clinical procedure (2) the ferric chloride-sulfuric acid solution is added to the solution of steroid in acetic acid. This step involves the liberation of considerable heat; to reduce the variability of heat liberation in mixing, we have modified the test as follows: To 0.01-1.0 mg. steroid obtained from an ethanol solution evaporated to dryness in a test tube is added 1.0 ml. glacial acetic acid. After solution of the steroid is effected, 4.0 ml. of a previously prepared and cooled mixture, consisting of 0.5 ml. of a solution of 2.5 Gm. FeCls. 61120 in 25 ml. glacial acetic acid, 50 ml. 96% sulfuric acid and 50 ml. of glacial acetic acid, is added by underlaying and then mixing. Two hours after mixing, the solution is read in a spectrophotometer against a reagent blank. The steroids investigated were of a high degree of purity, unless otherwise indicated; those with hormonal properties were obtained

Mechanisms of the Liebermann-Burchard and Zak color reactions for cholesterol.
Correlation of SO2 and Fe2+ measurements with new spectral data indicates that the LiebermannBurchard (L-B) and Zak color reactions for cholesterol have similar oxidative mechanisms, each yielding, as oxidation products, a homologous series of conjugated cholestapolyenes. These studies further suggest that the colored species observed in these two systems are enylic carbonium ions formed by pro...
متن کاملCharacteristics of a 17-ketosteroid reaction.
An improved method for the determination of 17-ketosteroids is described, which involves an increase in sensitivity by the simple expedient of upsetting the equilibrium of the reaction in removing the products of reaction by precipitating the chromophore formed. This precipitation system appears to achieve about 1.6 times the color sensitivity for the same reaction carried out as a solution pro...
متن کاملThe Correlation between the Endometrial Integrins and Osteopontin Expression with Pinopodes Development in Ovariectomized Mice in Response to Exogenous Steroids Hormones
Background: The ovariectomized animals are good models to evaluate the effect of different steroid hormone treatments on implantation events and the pattern of integrin expression. Therefore, this study was performed to compare the expression of integrins and osteopontin (OPN) in correlation with pinopode development in ovariectomized mice endometrium which was subjected to steroid hormones. Me...
متن کاملCholesterolMethodologies : A Review Bennie Zak
The introduction of enzymes as reagents into the methodologies of cholesterol determination (1-6) has drastically altered the limited strong-acid approaches taken by most analysts of the past 90 years (7-19). There has been a considerably heightened interest in determining the compound because of new diagnostic approaches such as hyperlipoprotein phenotyping (13) with the implied relationship o...
متن کاملProcedurefor SerumCholesterol, which Permitsthe QuantitativeEstimationofCholesterol Esters
The widely used procedure and manifold described in AutoAnalyzer Methodology N-24a (1) for serum total cholesterol yields generally lower results than those obtained by the manual procedure of Zak (i), on which it is based. Many laboratories have experienced this problem and some have adopted the questionable practice of standardizing the automated procedure against a commercial or previously a...
