Clinical and Population Studies Peripheral Artery Disease Is Associated With a High CD163/TWEAK Plasma Ratio

چکیده
Objective—In addition to its role in the clearance of haptoglobin-hemoglobin (Hp-Hb) complexes, CD163 is a macrophage scavenger receptor for tumor necrosis factor–like weak inducer of apoptosis (TWEAK). We recently reported that the CD163/TWEAK plasma ratio could be a potential biomarker of atherothrombosis in asymptomatic subjects. In this study, we assessed soluble TWEAK (sTWEAK) and soluble CD163 (sCD163) plasma levels in white males with peripheral artery disease (PAD) and in atherothrombotic femoral plaques to evaluate their relationship with disease. We also analyzed whether Hp-Hb complexes could compete for CD163-mediated TWEAK uptake. Methods and Results—Patients with PAD (n 155) showed a trend toward lower sTWEAK (median [interquartile range]: 134 [110–204] versus 147 [119–205] pg/mL; P 0.067) and higher sCD163 (median [interquartile range]: 367 [269–506] versus 288 [234–369] ng/mL; P 0.001) plasma concentrations than age-matched controls (n 251). sCD163 and sTWEAK plasma levels were negatively correlated in both patients and controls. After stratification according to the severity of disease, sCD163/sTWEAK ratio was significantly increased in patients with more severe disease relative to the other groups (P 0.049). Analysis of conditioned medium obtained from cultured human atherothrombotic femoral plaque samples (n 36) and healthy aortas (n 14) revealed that high amounts of sCD163 were released by the atherothrombotic tissue, whereas sTWEAK presented the opposite trend (P 0.05). Finally, we report a potential association between CD163 shedding and oxidative stress. Conclusion—Our results suggest that the sCD163/sTWEAK plasma ratio may be associated with atherothrombosis burden in PAD. We hypothesize that an imbalance between TWEAK and CD163 could reflect the progression of atherothrombosis. (Arterioscler Thromb Vasc Biol. 2010;30:1253-1262.)

Background: Differences in the prevalence and impact of hyperhomocytenemia on vascular disease between countries and races have been reported. Most studies have been undertaken in North American and European populations and the importance of plasma total homocysteine (tHcy) level as a risk factor for coronary artery disease in the Middle East particularly in Iran, however, is not known. متن کاملHigh serum sCD163/sTWEAK ratio is associated with lower risk of digital ulcers but more severe skin disease in patients with systemic sclerosis
INTRODUCTION Systemic sclerosis (SSc) is an autoimmune disease characterized by chronic inflammation, vascular injury and excessive fibrosis. CD163 is a scavenger receptor which affects inflammatory response and may contribute to connective tissue remodelling. It has recently been demonstrated that CD163 can bind and neutralize the TNF-like weak inducer of apoptosis (TWEAK), a multifunctional c...
متن کاملP178: Can Human T-Lymphotropic Virus Proviral Load Predict the Severity of Clinical Features in HAM/TSP Patients?
HTLV-1 is the causative agent for a neurologic disease named HTLV-I- associated myelopathy/tropical spastic paraparesis (HAM/TSP). Paraparesis of the lower limbs which appears gradually is the most common clinical feature of this disease. The indirect involvement of the nervous system by lymphocytes is more probable than the direct attack of the virus to the neurons. The proviral load (PVL) is ...
متن کاملPeripheral Eosinophilia and History of Allergy in Patients with Coronary Artery Disease
Background: Coronary artery diseases (CAD) is the most common cause of death in the developing counteries. Atherosclerosis is the leading cause of CAD. A number of studies have shown that total leukocyte count is a risk factor for CAD but there is little information on the role of eosinophilia and allergy. The aim of this study was to determine the relationship between eosinophilia and histor...
