چکیده

Beta (β)-Glucans are biological response modifiers with potent effects on the immune system. They are glucose polymers present in the cell walls of plants, fungi, and bacteria, and are considered to be pathogen-associated molecular patterns (PAMPs). It has been reported that these polymers have a variety of effects on the immune system, including anti-tumor and anti-infective activities against fungal, bacterial, viral, and protozoal infections. In this study, we investigated the effects of Saccharomyces cerevisiae β-glucan on IFN-γ production capacity of T cells. After 6 days incubation of human peripheral blood mononuclear cells in the presence or absence β-glucan, IFN-γ production and secretion were determined by flow cytometry and ELISA, respectively. β-glucan significantly upregulated IFN-γ production. In contrast, neutralization of IL-12 activity by anti-IL-12 decreased IFN-γ synthesis. These data suggest that β-glucan may support anti-tumour and anti-infective immune responses by increasing IL-12-induced IFN-γ production in T cells. Immunomodulatory activities associated with beta-glucan derived from Saccharomyces cerevisiae.

In this study we investigated the effect of beta-glucan derived from Saccharomyces cerevisiae on fungicidal activity, cytokine production and natural killer activity. Spleen and peritoneal cells from female C57BL/6 mice, previously injected (24 or 48 h) with 20 or 100 microg of glucan by i.p. route, were assayed. In vivo beta-glucan administration primed spleen cells for a higher production of ...

Human dendritic cell maturation and activation by a heat-killed recombinant yeast (Saccharomyces cerevisiae) vector encoding carcinoembryonic antigen.

Tumor-associated antigens are weakly immunogenic. Human carcinoembryonic antigen (CEA) is overexpressed on a wide range of human carcinomas and represents an attractive target for cancer immunotherapy. This study analyzes the ability of a Saccharomyces cerevisiae vector containing the transgene encoding CEA (yeast-CEA) to activate human dendritic cells (DCs) and stimulate CEA-specific T-cell re...

Glucan phosphate potentiates endotoxin-induced interferon-gamma expression in immunocompetent mice, but attenuates induction of endotoxin tolerance.

Glucan phosphate has been shown to enhance antimicrobial immunity in a variety of experimental models. However, the mechanisms by which glucans enhance resistance to infection remain largely unknown. Interferon-gamma (IFN-gamma) is a key regulator of both innate and acquired immunity. Suppression of IFN-gamma production is a prominent feature of the altered immune response that follows major tr...

Differential regulation of interleukin 12 and interleukin 23 production in human dendritic cells

We analyzed interleukin (IL) 12 and IL-23 production by monocyte-derived dendritic cells (mono-DCs). Mycobacterium tuberculosis H37Rv and zymosan preferentially induced IL-23. IL-23 but not IL-12 was efficiently induced by the combination of nucleotide-binding oligodimerization domain and Toll-like receptor (TLR) 2 ligands, which mimics activation by M. tuberculosis, or by the human dectin-1 li...

Cytokine-induced apoptosis of human natural killer cells identifies a novel mechanism to regulate the innate immune response.

Interferon-gamma (IFN-gamma) is critical for an effective innate immune response against infection. A combination of interleukins (ILs) derived from activated T cells (IL-2) and monocytes (IL-12), or monocytes alone (IL-15 and IL-12), induces optimal production of IFN-gamma from natural killer (NK) cells. The mechanism by which human NK cells downregulate their production of IFN-gamma is unknow...
