چکیده

Thyroid-stimulating hormone (TSH) increased within 1 mm the adenyl cyclase activity of bovine and canine thyroid homogenate. A similar effect was also found when adenyl cyclase activity was measured in thyroid slices. In experiments in vivo, adenyl cyclase activity was augmented within 3 min after TSH injection, but this effect did not persist for 60 min after the injection. Despite the transient stimulation of adenyl cyclase activity, increased 14C-l-glucose oxidation was still present 60 min after TSH injection. Stimulation in vitro of adenyl cyclase activity in thyroid homogenate was obtained using as little as 1 milliunit of TSH while adrenocorticotropic hormone and prolactin were without effect. TSH did not increase adenyl cyclase activity in adrenal or testis homogenates. TSH stimulation could still be obtained in the presence of 10m2 M NaF. TSH did not modify thyroid phosphodiesterase activity under conditions in which it stimulated adenyl cyclase activity. TSH rapidly increased cyclic adenosine 3’,5’-monophosphate (cyclic AMP) concentration in canine thyroid slices while luteinizmg hormone, adrenocorticotropic hormone, and prolactin were ineffective. Although NaF stimulated adenyl cyclase activity and 14C-l-glucose oxidation in canine thyroid slices, it did not increase cyclic AMP concentrations. During 15min incubations, lop2 M theophylline had very little effect on cyclic AMP levels, but significantly increased them during e-hour incubations. At both times, the effect of TSH on cyclic AMP was markedly potentiated by theophylline. Prostaglandin Ez reproduced effects of TSH on both l4C1 -glucose oxidation and cyclic AMP concentration in canine thyroid slices. Prostaglandin Fi, stimulated 14C-lglucose oxidation, but did not increase cyclic AMP levels. Prostaglandin B1 did not modify either cyclic AMP concentrations or W-l-glucose oxidation.

Effects of thyroid-stimulating hormone on adenyl cyclase activity and intermediary metabolism of "cold" thyroid nodules and normal human thyroid tissue.

"Cold" thyroid nodules do not concentrate (131)I before or after thyrotropin (TSH) administration. In an attempt to elucidate the reason for this TSH unresponsiveness, the effect of TSH in vitro on several metabolic parameters was studied in 11 "cold" thyroid adenomas, 2 medullary carcinomas, and in the surrounding normal thyroid tissue. Basal adenyl cyclase activity, glucose-1-(14)C oxidation,...

Adenyl Cyclase and Cyclic AMP Biochemical Links in the Regulation of Myocardial

The actions of many hormones may be related to their capacity to increase adenyl cyclase activity in their target organs. The evidence indicating that catecholamines and glucagon augment myocardial contractility by enhancing the activity of myocardial adenyl cyclase is summarized. Furthermore, data are presented suggesting that the inotropic actions of epinephrine and glucagon ultimately may be...

Thyrotropin increases prostaglandin levels in isolated thyroid cells.

We have shown that two unrelated prostaglandin antagonists block both thyrotropin (TSH) and prostaglandins E (PGE(1), PGE(2)) stimulation of thyroidal adenyl cyclase activation and cyclic 3',5'-adenosine monophosphate (cAMP) formation, suggesting that prostaglandins play an important role in regulating thyroid function. To further explore this postulate, we measured prostaglandin content by rad...
