چکیده
N-Acetylmuramyi-L-alanyl-D-isoglutamine (muramyl dipeptide, MDP) 1 was shown (1) to be the minimal and essential s t ructure requi red for adjuvant activity o f bacterial cell wall peptidoglycans. Fur ther , MDP has been found to possess various biological activities, o ther than adjuvant activity, that are exer ted by bacterial cell wall peptidoglycans. Since MDP is such a potent immunomodula tor , efforts are being made to apply it to a clinical use. MDP should be carefully used, however, because it can induce some detr imental side effects; MDP induced adjuvant arthritis in rats (2), enhanced endotoxic shock in guinea pigs (3), caused pyrexia and leucopenia in rabbits (4-6), and induced the lethargy in cats, rabbits, and guinea pigs (7, 8). We repor t here ano the r kind of undesired effect o f MDP. In the course of study of antigenicity of MDP, we found that MDP could cause severe necrotic inflammation. Heat-killed tubercle bacilli incorpora ted into Freund-type wateroil emulsion injected into guinea pig footpads induced extensive necrosis in the footpads at the site o f tubercle bacilli injection when, 3 -5 wk later, MDP dissolved in Dulbecco's phosphate-buffered saline (PBS) was injected intracutaneously at the flank, and some animals died f rom generalized shock.

Necrotic inflammatory reaction induced by muramyl dipeptide in guinea pigs sensitized by tubercle bacilli
In the course of studies aimed at determining whether MDP was antigenic or not, a hitherto unreported phenomenon was noticed. Injection (a provocative injection) of muramyl dipeptide (MDP) caused severe inflammation, with hemorrhage and necrosis in the footpads of guinea pigs, where tubercle bacilli in water-oil emulsion (a preparatory injection) had been injected 3-8 wk earlier. Sometimes the ...

متن کاملThe immune reaction against allogeneic necrotic cells is reduced in Annexin A5 knock out mice whose macrophages display an anti-inflammatory phenotype
Proteins of the annexin family bind to phospholipids in a Ca(2+) dependent manner. The exposure of phosphatidylserine (PS) by apoptotic as well as necrotic cells is one major eat-me-signal for macrophages. Annexin A5 (Anx A5) preferentially binds to PS. The availability of Anx A5 knock out (KO) mice allowed us to investigate for the first time if endogenous Anx A5 modulates the immune response ...
متن کاملSynergism between muramyl dipeptide and lipopolysaccharide in the inhibition of glycosaminoglycan synthesis in cultured rat costal chondrocytes.
The effect of synthetic muramyl dipeptide on glycosaminoglycan synthesis in cultured rat costal chondrocytes was examined. Muramyl dipeptide alone had no effect on the glycosaminoglycan synthesis of rat chondrocytes, whereas Escherichia coli lipopolysaccharide and interleukin 1 alpha inhibited glycosaminoglycan synthesis in a dose dependent manner. Muramyl dipeptide, when added to chondrocyte c...
متن کاملReversal of the vancomycin inhibition of peptidoglycan synthesis by cell walls.
Addition of cell walls to the peptidoglycan synthetase-acceptor system containing vancomycin (50 mug/ml) prevented the inhibition by the antibiotic. In addition, the inhibition of incorporation of [(14)C]muramyl-pentapeptide into peptidoglycan in the presence of vancomycin was reversed by the addition of cell walls to the assay mixture at 60 min. Cell walls previously saturated with vancomycin ...
