Use of micromethod for phenylalanine in management of phenylketonuric patients.

چکیده
FREATMENT of phenylketonuric infants with low phenylalanine diet requires frequent measurements of blood phenylalanine, since diet must be adjusted to changing phenylalanine requirements. The amount of blood required for repetitive determinations by the usual methods may be excessive. The need for frequent and rapid determinations led us to modify the method for serum phenylalanine (1) to permit use of blood obtained from a finger puncture-a modification suggested by methods for microchemical analysis for other blood constituents (2).

Biopterin derivatives in normal and phenylketonuric patients after oral loads of L-phenylalanine, L-tyrosine, and L-tryptophan.
Plasma biopterin derivatives studied in 10 normal and 21 phenylketonuric children showed a significantly high concentration in the latter group. Biopterin derivatives correlated with plasma phenylalanine concentration, but in normal adults given an oral phenylalanine load the rate of increase with phenylalanine differed from that in phenylketonuric patients. A patient with hyperphenylalaninaemi...

McKean, C. M. (1971). Archives of Disease in Childhood, 46, 608. Effects of totally synthetic, low phenylalanine diet on adolescent phenylketonuric patients. The long-term responses of 5 adolescent phenylketonuric patients to chemically-defined, synthetic diets with normal and low phenylalanine content were determined. The synthetic preparations were found capable of sustaining good health and ...
متن کاملDetection of phenylketonuric heterozygotes.
Estimation of plasma phenylalanine and tyrosine by an abridged (30 mm) column chromatography procedure was assessed. Plasma phenylalanine and tyrosine concentrations, determined in 112 phenylketonuric obligate heterozygotes and in 88 normal controls, did not differ significantly from other reported series in which the data were obtained by column chromatography. All these series were combined a...
متن کاملAspartame metabolism in normal adults, phenylketonuric heterozygotes, and diabetic subjects.
This study reviews clinical studies testing the effects of various doses of aspartame on blood levels of phenylalanine, aspartate, and methanol in normal subjects and known phenylketonuric heterozygotes. The effect of aspartame on the phenylalanine-to-large neutral amino acid ratio under various feeding situations is shown. The clinical studies of aspartame in diabetic subjects are limited to o...
متن کاملA model of human phenylalanine metabolism in normal subjects and in phenylketonuric patients.
The derivation of a quantitative model of phenylalanine metabolism in humans is described. The model is based on the kinetic properties of pure recombinant human phenylalanine hydroxylase and on estimates of the in vivo rates of phenylalanine transamination and protein degradation. Calculated values for the steady-state concentration of blood phenylalanine, rate of clearance of phenylalanine fr...
