چکیده

In the course of a series of investigations undertaken in this laboratory to obtain as complete a knowledge as possible of the physiology of normal and pathological joints, investigations concerning the metabolism of ‘synovial fluid mucin were undertaken. The two possible mechanisms for removal of mucin from the joint appear to be enzymatic destruction within the joint or removal by way of the lymphatics. In view of the known difficulty in removing globulin (a smaller molecule than mucin) from the joint (l), it seemed probable that enzymatic activity was the important factor. In an attempt to obtain evidence of the presence in normal or pathological fluid of an enzyme capable of destroying mucin, specimens of sterile normal and pathological fluids were allowed to stand for varying periods of time at 25’ and 37”. In no instance was any decrease in mucin noted nor did any change in mucin occur. However, in the course of other studies on synovial fluid mucin, a specimen of normal synovial fluid was encountered which had lost its viscosity and from which mucin could no longer be precipitated as a ropy mass by the addition of dilute acetic acid. The possibility that the mucin had been destroyed by an enzyme,

Studies on diffusing factors: The action of testicular extracts on the viscosity of vitreous humour preparations.

CHAIN & DUTHIE [1939] have recently shown that testicular extracts contain an enzyme capable of hydrolysing the mucins of synovial fluid and vitreous humour. Mucinases of a similar type have been obtained from pneumococci [Dubos, 1937; Meyer et al. 1937] and from haemolytic streptococci [Meyer, 1938]. Since the first visible effect of the testicular enzyme on the mucins examined was to reduce t...

Adhesion of Yersinia enterocolitica to purified rabbit and human intestinal mucin.

Interactions between Yersinia enterocolitica and purified intestinal mucins from rabbit and humans were investigated. Plasmid-bearing virulent organisms (but not plasmid-free nonvirulent bacteria) bound well to both mucins, suggesting that adherence was controlled by the virulence plasmid. Examination of binding to 14 different preparations of purified human intestinal mucin (8 preparations obt...

Divergent mechanisms of interaction of Helicobacter pylori and Campylobacter jejuni with mucus and mucins.

Helicobacter pylori and Campylobacter jejuni colonize the stomach and intestinal mucus, respectively. Using a combination of mucus-secreting cells, purified mucins, and a novel mucin microarray platform, we examined the interactions of these two organisms with mucus and mucins. H. pylori and C. jejuni bound to distinctly different mucins. C. jejuni displayed a striking tropism for chicken gastr...

The oligomeric structure of rat and human gastric mucins.

Intact oligomeric gastric mucins were isolated from the fundic part of rat and human stomach. Physicochemical properties of the oligomeric mucins from both species, such as buoyant density, molecular mass, proteinase-resistance, amino acid composition and monosaccharide composition were similar. Biochemical analysis showed that the oligomeric mucins from both species consist of disulphide-linke...

Sialomucin complex at the rat ocular surface: a new model for ocular surface protection.

The ocular surface, which is among the most accessible and vulnerable tissues in mammals, is protected by a complex tear film composed of lipid, aqueous and mucin layers. In spite of its importance, the molecular nature of the mucin contribution remains uncertain. Since membrane mucins have been implicated in the protection of other epithelia, we have analysed rat corneal and conjunctival tissu...
