Loss of hematopoietic progenitor cells CFU-GEMM, BFU-E, CFU-Mk, and CFU-GM in the acquired immunodeficiency syndrome (AIDS)?

چکیده
Hematological abnormalities have been found in the majority of patients with AIDS, in addition to immunological derangements [1]. Furthermore, treatment of opportunistic infections with folic acid antagonists and of neoplasms with cytostatic drugs is frequently complicated by profound thrombocytopenia, neutropenia, and anemia, thus resulting in the withdrawal of the drugs. In the present study we therefore analyzed the hematopoietic progenitor cell compartments, using an in vitro culture system that allows colony formation by the pluripotent progenitor cells CFU-GEMM and by the unipotent erythroid progenitors BFU -E, the megakaryocytic progenitors CFU-Mk, and the granulocytic-monocytic progenitors CFU-GM [2].

Effects of Recombinant a and ‘y Interferons on the In Vitro Growth of Circulating Hematopoietic Progenitor Cells (CFU-GEMM, CFU-Mk, BFU-E, and CFU-GM) From Patients With Myelofibrosis With Myeloid Metaplasia
Myeloflbrosis with myeloid metaplasia (MMM) is a chronic myeloproliferative disorder due to clonal expansion of a pluripotent hematopoietic progenitor cell with secondary marrow fibrosis. No definitive treatment has as yet been devised for this condition, which shows a marked variability in clinical course. To evaluate whether excessive hematopoietic progenitor cell proliferation could be contr...
متن کاملLack of evidence for infection of or effect on growth of hematopoietic progenitor cells after in vivo or in vitro exposure to human immunodeficiency virus.
The pathogenesis of the hematologic abnormalities commonly observed in patients with acquired immunodeficiency syndrome (AIDS) is incompletely understood. We report here that in vitro growth of myeloid (CFU-GM) and erythroid (BFU-E) progenitor cells from six patients with AIDS was not significantly different from that of normal human immunodeficiency virus (HIV) seronegative donors: 25.3 +/- 5 ...
متن کاملSynergistic Antiproliferative Effect of Recombinant Interferon-’y With Recombinant Interferon-a on Chronic Myelogenous Leukemia Hematopoietic
Recombinant interferons, a (rlFN-a) and ‘y (rIFN-’y), have been demonstrated to have significant antitumor activity as single agents in the treatment of chronic myelogenous leukemia (CML). Due to their possible synergistic efficacy. a combination rIFN therapy in CML has been proposed. To establish a biologic basis for this, we have studied the suppressive effects of rIFN-a and rlFN-’y on the in...
متن کاملEffect of recombinant interferons alpha and gamma on human bone marrow-derived megakaryocytic progenitor cells.
Interferons (IFNs) have been shown to suppress the proliferation of human pluripotent hematopoietic progenitor cells, CFU-GEMM, and committed erythroid (BFU-E, CFU-E) and granulocyte-macrophage (CFU-GM) progenitor cells. However, no information is yet available concerning the effect of IFNs on human megakaryocytic progenitor cells CFU-Mk. Furthermore the mechanisms underlying the inhibitory act...
متن کاملPreservation of Differentiation and Clonogenic Potential of Human Hematopoietic Stem and Progenitor Cells during Lyophilization and Ambient Storage
Progenitor cell therapies show great promise, but their potential for clinical applications requires improved storage and transportation. Desiccated cells stored at ambient temperature would provide economic and practical advantages over approaches employing cell freezing and subzero temperature storage. The objectives of this study were to assess a method for loading the stabilizing sugar, tre...
