چکیده

Changes in the activity of the tRNA methyltransferases have been found in all differentiating systems studied. Activity was examined in extracts of Rana pipiens embryos and in larval and adult liver by in vitro assay using S-adenosyl-L-[methyl-‘4C]methionine as the methyl donor. Specific activities of tRNA methyltransferases decreased, beginning with the time of feeding, when using high concentrations of the crude liver enzyme. A new methyltransferase activity, glycine Nmethyltransferase, appeared at the time of feeding. Apparently, the glycine methyltransferase is active before the onset of any of the characteristic metamorphic changes of other liver enzymes. Using partially purified enzyme from adult liver, the K, of glycine methyltransferase for Sadenosylmethionine is 0.3 mM and the K, for S-adenosylhomocysteine, a competitive inhibitor, is 0.08 mM.

Alteration of glycine N-methyltransferase activity in fetal, adult, and tumor tissues.

Glycine N-methyltransferase activity has been examined in a number of fetal and adult organs, as well as in several rodent hepatomas, using both enzymatic and immunological techniques. In fetal rabbit liver, the activity first appears at a low level at about 20 days postfertilization and rises to high levels after birth, reaching maximum in the adult liver. In fast-growing hepatomas, the activi...

O-11: N-a-acetyltransferase 10 Protein Regulates DNA Methylation and Embryonic Development

Background Genomic imprinting is a heritable and developmentally essential phenomenon by which gene expression occurs in an allele-specific manner1. While the imprinted alleles are primarily silenced by DNA methylation, it remains largely unknown how methylation is targeted to imprinting control region (ICR), also called differentially methylated region (DMR), and maintained. Here we show that ...

Convergent Mechanistic Features between the Structurally Diverse N- and O-Methyltransferases: Glycine N-Methyltransferase and Catechol O-Methyltransferase

Although an enormous and still growing number of biologically diverse methyltransferases have been reported and identified, a comprehensive understanding of the enzymatic methyl transfer mechanism is still lacking. Glycine N-methyltransferase (GNMT), a member of the family that acts on small metabolites as the substrate, catalyzes methyl transfer from S-adenosyl-l-methionine (AdoMet) to glycine...

Expression of 4 Genes in Ocimum basilicum and their Relationship with Phenylpropanoids Content

Recent data showed that phenylpropanoid compound, methylchavicol is essential component of Iranian cultivars of basil. Studying their occurrence during development of plant may help to elucidate the role of phenylpropanoids in plant cell physiology. We followed the phenylpropanoids concentration and the expression of genes related to their biosynthesis during growth and development of two culti...

Modeling of Methyl Transfer Reactions in S-Adenosyl-L-Methionine Dependent Enzymes

A very important trend for studying biomolecules is computational chemistry. In particular, nowadays it is possible to use theoretical methods to figure out the catalytic mechanism of enzyme reactions. Quantum chemistry has become a powerful tool to achieve a description of biological processes in enzymes active sites and to model reaction mechanisms. The present thesis uses Density Functional ...
