چکیده

THE stimulation of bone resorption by parathyroid hormone can be quantitated in tissue cultures of embryonic bone [20], using paired limb bones from a single embryo in which radioactive calcium (45Ca) has previously been incorporated. The ratio of 45Ca release from the parathyroid hormonetreated bones to that from the control bones is a sensitive measure of parathyroid effect. This technique has been applied to a study of the mechanism of action of parathyroid hormone and of other factors affecting bone resorption and also has been used to assay for parathyroid hormone activity in biological fluids [S, 21-231. The major disadvantages of this system are the variability of the response [al] and the difficulty in analyzing for small biochemical changes. Because our preliminary observations suggested that some of these disadvantages might be ascribable to the high concentration of serum (50 per cent) used in the culture medium, the objective of the present study was to find the simplest and chemically best defined medium in which bone resorption could be stimulated by parathyroid hormone. Chemically defined media without serum proved to be inadequate to support parathyroid hormone-induced bone resorption; therefore the minimum serum concentration required for a parathyroid effect was determined and the constituent of serum necessary for this response identified. The critical factor occurs in the albumin fraction of serum. This fraction appears to contain a non-parathyroid factor which can both support parathyroid-stimulated bone resorption and enhance 45Ca release from control bones.

The effect of a resistance training course with vitamin D supplementation on bone resorption markers in girls

Decreased bone density, especially in women, is a health risk factor, so the aim of this study was to investigate the effect of a resistance training course with vitamin D supplementation on bone Absorption markers in girls. Methods: The subjects of this semi experimental study were 24 non-athlete girls in Boroujerd (mean age 21.6 ± 2.04 years, mean height 155 ± 5.5 cm and average weight 64.13 ...

Effects of Oral Isotretinoin on Serum Vitamin D Metabolites and Other Biochemical Markers of Bone Turnover and Calcium Homeostasis in Severe Acne

Background: Few studies have investigated on vitamin D metabolites Serum levels, and calcium homeostasis in humans receiving retinoids, despite a substantial amount of literature concerning retinoid-induced osteoporosis in animals. We prospectively measured vitamin D metabolites serum levels and calcium homeostasis and radiographic bone changes in short course treatment with oral isotreti...

A role for interleukin-6 in parathyroid hormone-induced bone resorption in vivo.

Parathyroid hormone (PTH) exerts its regulatory effects on calcium homeostasis in part by stimulating the release of calcium from the skeleton. PTH stimulates bone resorption indirectly, by inducing the production by stromal/osteoblastic cells of paracrine agents which recruit and activate the bone-resorbing cell, the osteoclast. The identity of the stromal cell/osteoblast-derived paracrine fac...

P-181: Protective Role of Vitamin E As An Alternative Treatment for Ovariectomized Osteoporotic Rats

Background: Osteoporosis one of the postmenopausal symptoms is characterized by bone loss. There is a link between excessive reactive oxygen species (ROS) formation, estrogen deficiency due to cessation of ovarian function and bone loss. Free radicals are responsible for causing osteoblast apoptosis and reducing osteoblastogenesis in bone remodeling. Vitamin E is a potent antioxidant with the a...

Direct extraction and assay of bone tissue collagenase and its relation to parathyroid-hormone-induced bone resorption.

A method has been developed for the quantitative extraction of collagenase from as little as one 19-day-fetal-mouse calvarium. About 20-40 munits of collagenase are extracted per mg of tissue, all in a latent form that, after proper activation, shows the typical properties of mammalian collagenase. Culturing the calvaria for 2 days with parathyroid hormone (PTH) increases their procollagenase c...
