چکیده

In this study, two novel boron dipyrromethene-based photosensitizers (BDP3 and BDP6) substituted with three or six trifluoromethyl groups have been synthesized and characterized with various spectroscopic methods, and their photo-physical, photo-chemical, and photo-biological properties have also been explored. The two photosensitizers are highly soluble and remain nonaggregated in N,N-dimethylformamide as shown by the intense and sharp Q-band absorption. Under red light irradiation (λ = 660 nm, 1.5 J/cm²), both photosensitizers show high and comparable cytotoxicity towards HepG2 human hepatocarcinoma and HeLa human cervical carcinoma cells with IC50 values of 0.42-0.49 μM. The high photocytotoxicity of BDP3 and BDP6 can be due to their high cellular uptake and low aggregation tendency in biological media, which result in a high efficiency to generate reactive oxygen species inside the cells. Confocal laser fluorescence microscopic studies indicate that they have superior selective affinities to the mitochondria and lysosomes of HepG2 and HeLa cells. The results show that these two trifluoromethyl boron dipyrromethene derivatives are potential anticancer agents for photodynamic therapy.

In vitro and in vivo photocytotoxicity of boron dipyrromethene derivatives for photodynamic therapy.

To understand the effects of substitution patterns on photosensitizing the ability of boron dipyrromethene (BODIPY), two structural variations that either investigate the effectiveness of various iodinated derivatives to maximize the "heavy atom effect" or focus on the effect of extended conjugation at the 4-pyrrolic position to red-shift their activation wavelengths were investigated. Compound...

BODIPY-doped silica nanoparticles with reduced dye leakage and enhanced singlet oxygen generation

Photodynamic therapy (PDT) is a promising modality for cancer treatment. The essential element in PDT is the photosensitizer, which can be excited by light of a specific wavelength to generate cytotoxic oxygen species (ROS) capable of killing tumor cells. The effectiveness of PDT is limited in part by the low yield of ROS from existing photosensitizers and the unwanted side effects induced by t...

Nanomaterials and singlet oxygen photosensitizers: potential applications in photodynamic therapy

In this review, we address a highly interdisciplinary field: the use of nanomaterials as carriers for singlet oxygen photosensitizers and their potential applications in photodynamic therapy. In particular, recent advances in the use of nanoparticles including inorganic oxide-, metallic-, and polymer-based nanocomposites as photosensitizer carriers are highlighted. We review advantages and shor...

بررسی اثر فتودینامیکی کمپلکس تتراپیریدینو پورفیرازین روی (II)، بر رده‌ی سلولی HeLa

Background and Objective: Photodynamic therapy is a treatment that uses photosensitizer and intense visible light. When photosensitizers get exposed to a specific light wavelength (preferentially in the red region), they produce reactive oxygen species that are toxic to cells. Recently, attention has been focused on porphyrins and their analogs as photosensitizers. Zn (II) tetrapyridinoporphyra...

Photodynamic therapy (PDT)

Photodynamic therapy (PDT) is a comparatively novel therapeutic method involving a safe light source and a light-sensitive substance, termed as photosensitizers (PSs), such as methylene blue (MB), toluidine blue (TBO), sulfonated aluminum phthalocyanine, chlorine derivatives, nontoxic indocyanine green (ICG), and curcumin (CUR). (1) The combination of a nontoxic PS with low-intensity visible li...
