چکیده

Results Under basal conditions, the major prostaglandin released by human microvascular endothelial cells is PGI2. Exogenous PGI2 markedly attenuated the migration of isolated eosinophils through cell-free filters. This effect was prevented by the IP receptor antagonist Cay10441 and the adenylyl cyclase inhibitor SQ29548. Treatment of endothelial cells with diclofenac, a non-selective COX inhibitor, abolished the PGI2 production which was accompanied by enhanced eosinophil adhesion to endothelial monolayers. Similarly, the transmigration of eosinophils through endothelial monolayers was enhanced by diclofenac. The diclofenac treatment itself decreased the electrical resistance of endothelial monolayers and compromised the intercellular cell junctions as visualized by VE-cadherin staining.

تمایز سلول‌های دندریتیک مشتق از مونوسیت بر روی لایه‌ای از سلول‌های اندوتلیال به‌عنوان لایه تغذیه‌کننده

Background: The innate and adaptive immune responses are dependent on the migration of leukocytes across endothelial cells. Dendritic cells (DCs) play an important role in the initiation of cellular immune responses during their migration from tissues into the lymph nodes where they interact with endothelial cells of lymphatic vessels. We investigated the effect...

Endothelial prostaglandin I2 restrains eosinophil trafficking

Background Enhanced eosinophil extravasation into the tissue is a characteristic feature of bronchial asthma and other allergic diseases. The barrier-forming vascular endothelial cells release prostaglandin I2 (PGI2, prostacyclin) as the major prostanoid, and it has been previously observed that PGI2 receptor (IP)-deficient mice show enhanced eosinophilic inflammation in response to allergens. ...

Determination the Role of Endothelial Cell-Specific Molecule-1(ESM-1) in Childhood Bronchial Asthma

Background Endothelial Cell-Specific Molecule-1 (ESM-1) is a 50 kDa soluble proteoglycan that is produced mainly the vascular endothelial cells of the kidney and lung. It is produced by the effects of proangiogenic and pro-inflammatory cytokines, and it indicates activation and dysfunction of the vascular endothelium. We aimed to detect the role of ESM-1 in childre...

A study on the relationship between clinical features with Ki67 expression and eosinophil cells infiltration in oral squamous cell carcinoma

Background :Cell proliferation is one of the most critical factors in metastasis and prognosis of the malignant tumors.Recent investigations show that the eosinophil granolosytes have an important role in developing of malignant tumors. The relation between cell proliferation and eosinophilic infiltration in oral squamous cell carcinoma (OSCS) with prognosis is unclear. The aim of this study w...

Spironolactone Inhibits NADPH Oxidase-Mediated Oxidative Stress and Dysregulation of the Endothelial NO Synthase in Human Endothelial Cells

Accumulating evidence indicates that aldosterone plays a critical role in the mediation of oxidative stress and vascular damage. NADPH oxidase has been recognized as a major source of oxidative stress in vasculature. However, the relation between NADPH oxidase in aldosterone-mediated oxidative stress in endothelial cells remains to be ascertained. The present study aimed to investigate the rel...
