Studies on Non-precipitating Antibody Ii. Circulating Antibody-antigen Complex following Repeat Injections of Antigen*
نویسنده
چکیده
When a single intravenous injection of foreign protein is given to rabbits, large amounts of antibody-antigen complex appear in the circulation. The complex usually is first detected on the 7th day, at a time when free circulating antigen is still present. I t reaches a maximum on the 12th day, when free antigen disappears. After the 12th day antibody-antigen complex slowly dedines, while free precipitating ant ibody may be found in the circulation (1). I t appeared interesting to s tudy the formation of antibody-antigen complex after repeated injections of antigen and to correlate the data with the levels of free antigen and free precipitating ant ibody and with the appearance of free non-precipitating ant ibody (2). The present paper is a report of such studies.

In a previous paper (1) we described the appearance of a soluble circulating antibody-antigen complex in rabbits after single intravenous injections of large doses of bovine plasma albumin or bovine plasma gamma globulin, or after intramuscular injections of these agents with Freund's adjuvants. The soluble complex was demonstrated in the serum by brief exposure to alkali in the cold followed b...

Reinjection of antigen into immunized rabbits usually results in an immediate increase of circulating antibody-antigen complex. This immediate formation of complex seems to depend upon the presence of non-precipitating antibody before reinjection. The complex disappears by the 2nd day after reinjection and reappears between the 4th to 7th day. This is earlier than its first appearance after a p...

Background and objectives: Anti-human immunoglobulin G (IgG) antibody is produced against human IgG in various laboratory animals. The present study tried not only to produce anti-human IgG, but also to assess different antigen injection techniques leading to optimal production of anti-human globulin. Methods: The antibody was separated from human serum using precipitation method with sodium sul...
متن کاملProduction and Characterization of a Monoclonal Antibody against an Antigen on the Surface of Non-Small Cell Carcinoma of the Lung
Background: Lung carcinoma is a multiple type cancer comprising of small cell and non-small cell carcinomas (NSCLC). For therapeutic and diagnostic purposes, serum monoclonal antibodies have been produced against lung cancer. Objective: To charac-terize a murine monoclonal antibody (ME3D11) reactive with human NSCLC. Methods: A murine monoclonal antibody (ME3D11) reactive with human NSCLC was s...
متن کاملHeterologous Immunization, a Way out of the Problem of Monoclonal Antibody Production against Carcinoma Antigen 125
Background: Monoclonal antibodies (mAbs) are essential tools for many molecular im-munology investigations, epitope mapping and molecular modelling, clinical laboratory di-agnostic tests and immunotherapy. Humoral immune response of immunized animals largely depends on the nature of antigen and the immunization technique. Polysaccharides and heavily-glycosylated proteins are very elusive target...
