چکیده

Phage-encoded peptide libraries are often used to study biomolecular interactions. We employed this combinatorial approach to identify ligands specific for the streptococcal collagen-like proteins, Scl1 and Scl2, which are expressed on the cell surface by group A Streptococcus. Several sequence motifs displayed by phages were selected for their binding ability to different Scl variants. D 2005 Elsevier B.V. All rights reserved.

Unique Footprint in the scl1.3 Locus Affects Adhesion and Biofilm Formation of the Invasive M3-Type Group A Streptococcus

The streptococcal collagen-like proteins 1 and 2 (Scl1 and Scl2) are major surface adhesins that are ubiquitous among group A Streptococcus (GAS). Invasive M3-type strains, however, have evolved two unique conserved features in the scl1 locus: (i) an IS1548 element insertion in the scl1 promoter region and (ii) a nonsense mutation within the scl1 coding sequence. The scl1 transcript is drastica...

Harnessing the Versatility of Bacterial Collagen to Improve the Chondrogenic Potential of Porous Collagen Scaffolds

Collagen I foams are used in the clinic as scaffolds to promote articular cartilage repair as they provide a bioactive environment for cells with chondrogenic potential. However, collagen I as a base material does not allow for precise control over bioactivity. Alternatively, recombinant bacterial collagens can be used as "blank slate" collagen molecules to offer a versatile platform for incorp...

Engineered recombinant bacterial collagen as an alternative collagen-based biomaterial for tissue engineering

The key structural and signaling roles of collagen in the extracellular matrix (ECM) make it an attractive biomaterial for tissue engineering, but there are limitations in the standardization and purity of natural collagen sources currently available for such applications (Ruggiero and Koch, 2008; Werkmeister and Ramshaw, 2012). Significant effort has been made to produce human collagen in reco...

Mechanism of stabilization of a bacterial collagen triple helix in the absence of hydroxyproline.

The Streptococcus pyogenes cell-surface protein Scl2 contains a globular N-terminal domain and a collagen-like domain, (Gly-Xaa-X'aa)(79), which forms a triple helix with a thermal stability close to that seen for mammalian collagens. Hyp is a major contributor to triple-helix stability in animal collagens, but is not present in bacteria, which lack prolyl hydroxylase. To explore the basis of b...

Prediction of T-cell epitopes for designing a reverse vaccine against streptococcal bacteria

Streptococcal bacteria are among dangerous human pathogens with major prevalence worldwide. A good vaccine against streptococcal bacteria should have epitopes that confer protection from infection by different streptococcal bacteria types. we aimed was to recognize the most immunogenic and conserved epitopes of streptococcal bacteria, which could be a potential candidate for vaccine development...
