چکیده
In the brain, N-methyl-D-aspartate (NMDA)-type glutamate receptors are important elements for the manifestation of memory as well as mediators of neurotoxicity, and they are thought to be exclusive to neurons. To test for the expression of functional NMDA receptors on astrocytes, we generated transgenic mice in which glial fibrillary acidic protein (GFAP)-positive astrocytes are labeled by a green fluorescent protein and tested their responses to NMDA in acute cortical slices by patch-clamp recording and Ca imaging. The NMDA-evoked currents reversed at 0 mV; could be blocked by MK-801; persisted in the absence of synaptic transmission; were sensitive to Mg; and were accompanied by focal Ca elevation, indicating the presence of functional NMDA receptors. Furthermore, we detected mRNAs for NMDA receptor subunits in freshly isolated astrocytes purified by fluorescence-activated cell sorting. We conclude that processes of cortical astrocytes enwrapping synaptic regions express high densities of NMDA receptors that could be involved in neurone-glia signaling.

Distinct pharmacological and functional properties of NMDA receptors in mouse cortical astrocytes.
BACKGROUND AND PURPOSE Astrocytes of the mouse neocortex express functional NMDA receptors, which are not blocked by Mg(2+) ions. However, the pharmacological profile of glial NMDA receptors and their subunit composition is far from complete. EXPERIMENTAL APPROACH We tested the sensitivity of NMDA receptor-mediated currents to the novel GluN2C/D subunit-selective antagonist UBP141 in mouse co...
متن کاملEffects of spironolactone and fludrocortisone on neuronal and glial toxicity induced by N-methyl-D-Aspartate and chloroquine in cell culture
Spironolactone has produced beneficial effects in animal models of neurodegenerative disorders. However, the underlying mechanisms of this agent on neurons and glia are mostly unknown. Therefore, we aimed to show the effects of spironolactone and fludrocortisone, a mineralocorticosteroid receptor agonist, on neuronal and glial toxicity induced by N-methyl-D-aspartate (NMDA) activation and chlor...
متن کاملLocally synchronized astrocytes.
Astrocytes exhibit spontaneous calcium fluctuations. These activities have not been captured by large-scale recordings, and little is known about their collective dynamics. In situ and in vivo calcium imaging from hundreds (up to 2195) of astrocytes in the mouse hippocampus and neocortex revealed that neighboring astrocytes spontaneously exhibited synchronous calcium elevations and formed local...
متن کاملAmelioration of Pentylenetetrazole-Induced Seizures by Modulators of Sigma, N-Methyl-D-Aspartate, and Ryanodine Receptors in Mice
Background: Sigma receptors, N-methyl-D-aspartate (NMDA) antagonist, and modulators of intracellular calcium may be useful for seizure control. Therefore, we aimed to evaluate the antiepileptic effects of opipramol, a sigma receptor agonist, against pentylenetetrazole (PTZ)-induced seizures in mice and assess ketamine and caffeine interaction with the antiepileptic effects of opipramol.Methods:...
متن کاملCannabinoid receptors contribute to astroglial Ca2+-signalling and control of synaptic plasticity in the neocortex
Communication between neuronal and glial cells is thought to be very important for many brain functions. Acting via release of gliotransmitters, astrocytes can modulate synaptic strength. The mechanisms underlying ATP release from astrocytes remain uncertain with exocytosis being the most intriguing and debated pathway. We have demonstrated that ATP and d-serine can be released from cortical as...
